摘要
目的检测结肠癌组织中半胱氨酰白三烯受体-1(CysLTR-1)mRNA的表达,并探讨其意义。方法收集70例结肠癌患者的癌组织及其癌旁正常组织,采用Real-time PCR法检测各组织样本中CysLTR-1 mRNA相对表达量,分析CysLTR-1表达与患者临床病理特征的关系及CysLTR-1表达对结肠癌患者预后的影响。结果结肠癌组织中CysLTR-1 mRNA相对表达量高于癌旁正常组织,差异有统计学意义(P=0.000)。结肠癌组织中CysLTR-1表达与肿瘤大小、分化程度、淋巴结转移和TNM分期相关(P均<0.05)。Kaplan-Meier生存分析结果显示,CysLTR-1高表达者的术后3年总生存率低于CysLTR-1低表达者(P=0.032)、中位生存时间短于CysLTR-1低表达者(P=0.049)。结论结肠癌组织中CysLTR-1 mRNA呈高表达;监测结肠癌组织中CysLTR-1表达,有助于对患者进行病情判断和预后评估。
Objective To detect the expression of cysteinyl leukotriene receptor-1（ CysLTR-1） mRNA in the colon carcinoma tissues and to discuss its significance. Methods The cancer tissues and the adjacent normal tissues from 70 patients with the colon carcinoma were collected. The relative expression level of CysLTR-1 mRNA in each tissue sample was determined by real-time PCR. Meanwhile,we analyzed the correlations between the CysLTR-1 expression and the clinicopathological characteristics,and evaluated the effect of CysLTR-1 expression on the patients ＇ prognosis. Results The relative expression level of CysLTR-1 mRNA of the colon carcinoma tissues was much higher than that in the adjacent normal tissues,with statistically significant difference（ P = 0. 000）. In the colon carcinoma tissues,the CysLTR-1 expression was correlated with the tumor size,the differentiation degree,the lymph node metastasis,and the TNM stages（ all P〈0. 05）. The Kaplan-Meier results suggested that,the overall 3-year survival rate of patients with high CysLTR-1 expression was lower than that of patients with low CysLTR-1 expression（ P = 0. 032）,and the medium survival time was significantly shorter than that of patients with low CysLTR-1 expression（ P = 0. 049）. Conclusion CysLTR-1 mRNA is highly expressed in the colon cancer tissues,and monitoring the expression of CysLTR-1 in postoperation is helpful to evaluate the progression and prognosis of patients.
