卵清蛋白金纳米簇水浴合成及汞离子检测应用 被引量:2

摘要 以卵清蛋白为还原剂,通过水浴加热法,合成了具有红色荧光的卵清蛋白-金纳米簇。通过优化得到了反应最佳条件。考察了不同有机溶剂和金属离子对金纳米簇荧光的影响,并测定了检测汞离子的线性范围。试验结果表明,当氯金酸浓度固定为2.33mM,水浴加热温度固定在37℃时,反应最佳条件为卵清蛋白浓度10.71mg mL^(-1),NaOH浓度0.095M,反应时间6h。生成的金纳米簇在400~600nm范围内无明显紫外吸收峰,最大发射峰位于641nm。金纳米簇呈球形,颗粒大小约为2-3nm,荧光量子产率为0.0516。有机溶剂和金属离子对金纳米簇荧光均有一定影响,其中汞离子对金纳米簇荧光的影响最为显著,在5×10^(-7)-1×10^(-5)M浓度范围呈现良好的线性关系。试验结果表明,通过卵清蛋白合成的金纳米簇具有良好的荧光性能,且可用于水样中汞离子的检测。 In this paper,Ovalbumin-gold nanoclusters( OVA-AuNCs) with red fluorescence was synthesized by water-bath heating method using OVA as the reducing agent.Reaction conditions were optimized,and influences of different solvents or metal ions on the fluorescence intensity of the AuNCs were studied.When the concentration of HAuCl4 was constant at 2. 33 mM,the optimum reaction conditions were as follows: concentration of OVA and NaOH 10. 71 mg mL^(-1) and 0. 095 M,respectively,and the reaction time was 6 h.Synthesized AuNCs showed no obvious absorption peak in the range of 400-600 nm,and the maximum emission peak was641 nm.The diameter of the synthesized AuNCs was 2-3 nm with the spherical size,and the quantum yield was 0. 0516. Organic solvents and metal ions all showed some influence on the fluorescence intensity of the AuNCs,and mercury ions could quench the fluorescence intensity of AuNCs significantly.The fluorescence intensity of OVA-AuNCs toward mercury ions decreased linearly over the mercury ions concentration range of 5 × 10^(-7)-1× 10^(-5) M. Results showed that AuNCs synthesized by OVA showed good fluorescence and could be used in the determination of mercury ions in water samples.
