摘要
目的:探讨子痫前期胎盘组织中AP-2α与滋养细胞凋亡、侵袭的相关性。方法:选取2015年11月~2017年10月间在本院分娩的子痫前期产妇68例作为子痫前期组,另取同期在本院进行分娩的健康产妇60例作为正常对照组。对比两组胎盘组织中AP-2α基因、凋亡相关基因、侵袭相关基因表达量的差异,进一步分析子痫前期胎盘组织中AP-2α基因表达量与滋养细胞凋亡、侵袭活性的内在联系。结果:子痫前期胎盘组织中AP-2αmRNA的表达量高于正常对照组;凋亡相关基因Bcl、Cx43 mRNA的表达量低于正常对照组,Bax、Caspase7、metastin mRNA的表达量高于正常对照组;侵袭相关基因HIF-1α、GPR30、MEST、Snail mRNA的表达量低于正常对照组,PEDF、RECK mRNA的表达量高于正常对照组(P<0.05)。Pearson检验发现,子痫前期胎盘组织中AP-2α基因表达量与滋养细胞凋亡相关基因、侵袭相关基因表达量均直接相关。结论:子痫前期产妇胎盘组织中AP-2α基因表达量异常增高,可正向调控滋养细胞的凋亡活力并负向调控其侵袭活力。
Objective:To investigate the correlation of AP-2αwith trophoblast apoptosis and invasion in placental tissues of preeclampsia.Methods:68 puerperae with preeclampsia who gave birth in our hospital between November 2015 and October 2017 were selected as preeclampsia group,and 60 healthy puerperae who gave birth in our hospital during the same period were selected as normal control group.The differences in expression of AP-2αgene,apoptosis-related genes and invasion-related genes in placenta tissue were compared between the two groups,and the inner link of AP-2αgene expression with trophoblast apoptosis and invasion activity in placental tissues of preeclampsia was further analyzed.Results:AP-2αmRNA expression in placental tissue of preeclampsia group was higher than that of control group;apoptosis-related genes Bcl and Cx43 mRNA expression were lower than those of control group whereas Bax,Caspase7 and metastin mRNA expression were higher than those of control group;invasion-related genes HIF-1α,GPR30,MEST and Snail mRNA expression were lower than those of control group whereas PEDF and RECK mRNA expression were higher than those of control group(P〈0.05).Pearson test showed that the AP-2αgene expression in the placental tissues of preeclampsia was directly correlated with the expression of trophoblast apoptosis-related genes and invasion-related genes.Conclusion:AP-2αgene expression significantly increases in the placental tissue of puerperae with preeclampsia,and it can positively regulate the trophoblast apoptosis activity and negatively regulate its invasion activity.
