摘要
H3亚型流感病毒是威胁公共卫生安全以及动物生命安全的主要流感病毒亚型之一。为了解H3N7亚型流感病毒的生物学特性,本研究对2015年浙江省分离到的一株H3N7亚型禽流感病毒(AIV A/duck/Zhejiang/S1075/2015(H3N7)(简称DK/ZJ/S1075/2015)进行了遗传演化分析、抗原性分析以及小鼠感染性实验。遗传演化分析结果显示,该病毒株的聚合酶碱性蛋白2(PB2)基因、核蛋白(NP)基因、神经氨酸酶(NA)基因、非结构蛋白(NS)基因分别与H4N2、H7N3、H10N7、H4N6亚型流感病毒有着密切的关系,表明该株病毒的基因来源具有明显的遗传多样性。分离株的抗原性分析结果显示该病毒株与本实验室分离的多株H3亚型AIV存在16倍以上的抗原性差异,表明其发生了明显的抗原漂移。小鼠感染性实验结果表明,分离株无需提前适应即可在小鼠肺脏和鼻甲中复制,但小鼠感染病毒后无明显临床症状,与对照组相比其体重变化不明显,呈现低致病性。本研究结果为H3亚型流感病毒的全面监测和综合防控提供了数据支持。
H3 subtype influenza A virus is one of the main subtypes that threats both public and animal health. To investigate the biological characteristics of an H3N7 subtype avian influenza virus (AIV) A/duck/Zhejiang/S1075/2015(H3N7) isolated in Zhejiang province in 2015, the virus was characterized by phylogenetic analyses, antigenicity and BALB/c mice infection assays. Phylogenetic analyses of full genomes showed that the PB2, NP, NA and NS genes of the virus were most closely related to the H4N2, H7N3, H10N7, H4N6 subtype influenza viruses and the genetic origin of the virus was obviously diversified. There was more than 16-fold difference between the HI tites of the H3N7 AIV and laboratory-confirmed H3 subtype influenza viruses. Antigenicity assays of the H3N7 subtype AIV revealed that it had the phenomenon of antigenic drift. The H3N7 AIV was able to replicate in lung and nasal turbinate of infected mice without prior adaptation, which caused no obvious clinical symptoms or weight loss after infection, indicating the H3N7 subtype AIV was a low pathogenic virus to mice. Hence, it was significiant to improve the comprehensive survelliance strength and control of H3 subtype influenza virus.
