UPLC法同时测定四君子汤中7种化学成分的含量 被引量:4

Simultaneous determination of seven chemical constituents in Sijunzi Decoction by UPLC method

摘要 目的建立一种在双波长下同时测定四君子汤中人参皂苷Rg1、Re、Rf、Rb1、Rd、甘草酸和白术内酯Ⅲ7个化学成分含量的方法。方法采用超高效液相色谱法,Universil XB C8色谱柱(150 mm×2.1 mm,1.8μm),流动相:乙腈(A)-体积分数0.05%磷酸水溶液(B),梯度洗脱,流速0.2 m L·min^(-1),检测波长203 nm测定人参皂苷Rg1,Re,Rf,Rb1,Rd;220 nm测定甘草酸,白术内酯Ⅲ,进样量2μL,柱温为35℃。结果人参皂苷Rg1、Re、Rf、Rb1、Rd、甘草酸、白术内酯Ⅲ质量浓度分别在149.6~897.6 mg、100.2~601.2、49.2~295.2、150.4~902.4、80.2~481.2、240.6~1 443.6和15.8~94.8 mg·L^(-1)内与峰面积呈良好的线性关系,平均回收率分别为99.1%、99.5%、98.5%、98.9%、98.7%、99.3%和99.3%,RSD为1.3%、1.0%、1.5%、1.2%、1.2%、1.3%和1.0%(n=6)。结论本方法可用于四君子汤中人参皂苷Rg1、Re、Rf、Rb1、Rd,甘草酸、白术内酯Ⅲ的含量测定,为四君子汤质量控制提供依据。 Objective To establish a method for the simultaneous determination of ginsenoside Rg1,Re,Rf,Rb1,Rd,glycyrrhizic acid and albicolide III in four giants under dual wavelengths. Methods The UPLC separation was achieved on an Universil XB C8 column( 150 mm × 2. 1 mm,1. 8 μm) with 0. 05%phosphoric acid aqueous solution( A)-acetonitrile( B) as mobile phase at the flowrate of 0. 2 mL·min^(-1) for gradient elution. The column temperature was 35 ℃. The PDA detection wavelengths were set at 203 nm for ginsenoside Rg1,Re,Rf,Rb1 and Rd,220 nm for glycyrrhizic acid and Atractylodes Ⅲ,injection 2 μL.Results The calibration curve was linear within the range of 149. 6-897. 6 mg · L^-1,100. 2-601. 2 mg·L^-1,49. 2-295. 2 mg·L^-1,150. 4-902. 4 mg·L^-1,80. 2-481. 2 mg·L^-1,240. 6-1 443. 6 mg·L^-1 and 15. 8-94. 8 mg·L^-1 for ginsenoside Rg1,Re,Rf,Rb1,Rd,glycyrrhizic acid and albicin III,respectively. The average recoveries of the seven constituents were 99. 5%,98. 9%,98. 7%,99. 3% and 99. 3%,RSD were 1. 3%,1. 0%,1. 5%,1. 2%,1. 2%,1. 3% and 1. 0%( n = 6),respectively.Conclusion The method is simple and accurate,and it can be used for the determination of ginsenoside Rg1,Re,Rf,Rb1,Rd,glycyrrhizic acid and albicin III in Sijunzi Decoction,which provides the basis for the quality control of Sijunzi Decoction.
