响应面法优化罗布麻多糖提取工艺及抗氧化活性研究 被引量:14

摘要 研究罗布麻多糖最佳提取工艺及其抗氧化活性。在单因素试验的基础上,采用Box-Benhnke对影响提取罗布麻多糖得率的3个主要因素进行优化,并通过DPPH·清除率、·OH清除率和铜离子还原能力试验评价罗布麻多糖的体外抗氧化活性。通过单因素试验及Box-Benhnken试验设计可知,在提取温度91℃,提取时间1.75 h,料液比1∶82(g/m L)的条件下,罗布麻粗多糖的提取率为1.86%。随着样品浓度的增加,其抗氧化能力逐渐增强,其中对DPPH·的清除能力及对铜离子的还原能力较为显著,与天然抗氧化剂VC相近。 Study on optimum extraction technology and antioxidant activity of Apocynum venetum polysaccharides,based on single factor test,the three main factors affecting extraction of polysaccharide yield from Apocynum venetum were optimized by response surface methodology. The extraction technology was optimized by single factor and response surface analysis. The antioxidant activity of Apocynum venetum polysaccharides was evaluated by DPPH radical scavenging rate,OH radical scavenging rate and copper ion reduction ability. The optimal extraction technology was as follow:liquid to solid was 1 ∶ 82(g/m L),ultrasonic extraction time was1.75 h,extraction temperature was 91 ℃. The extraction rate of crude polysaccharide of Apocynum venetum was1.86 % With the increase of the concentration of the sample,its antioxidant ability gradually increased. Among them,the scavenging ability of DPPH·and the reduction ability of copper ion were more obvious,which was similar to that of natural antioxidant VC.
