摘要 评价氧多巴胺仿生修饰超顺磁性纳米颗粒(polymerization dopamine modified superparamagnetic iron oxide nanoparticles,PDA-SPIONs)作为体外基因载体的可行性.方法:利用琼脂糖凝胶电泳对PDA-SPIONs结合DNA能力进行表征,应用绿色荧光蛋白报告基因系统,检测PDA-SPIONs将外源基因转染至HEK293T细胞系的转染可能性.结果:PDA-SPIONs对HEK293T细胞系形态产生明显影响,将外源基因转染至HEK293T细胞系的能力较弱.结论:PDA修饰的磁性纳米颗粒需进一步修饰改造才能用于体外转染的新型非病毒基因载体. AIM: To evaluate the feasibility of biomimetic modified superparamagnetic iron oxide nanopar ticles (PDA SPIONs) as gene vector on efficiency of transfection. METHODS: The potential of adsor bing DNA of PDA SPIONs was analyzed by electrophoresis. Transfection was determined by delivering reporter gene Green fluorescent protein (GFP) control encoding, to HEK293 and A549 cell lines using PDA SPIONs as vector. The Lipofectamine 2000 transfection kit was used as the positive control. RE SULTS.- Foreign gene could be delivered to HEK293 but not A549 cell lines by Lipofectamine 2000 and ex pressed with high efficiency, but PDA SPIONs shows very low transfection efficiency in the HEK293 cell lines. Magnetic field or other advanced modification maybe needed to enhance the efficiency of transfec tion. CONCLUSION:PDA SPIONs should be further modified for using as a non viral gene vector in vitro.
