益肺逐积方联合环磷酰胺对Lewis肺癌小鼠肿瘤组织CD151、整合素α_3及β_1 mRNA表达水平的影响 被引量:1

Effect of Yifei Zhuji Formula Combined with Cyclophosphamide on CD151,ITG α_3 and ITG β_1 mRNA Expression in Tumor of Mice with Lewis Lung Cancer

摘要 目的研究益肺逐积方联合环磷酰胺(cyclophosphamide,CTX)对Lewis肺癌小鼠肿瘤组织CD151、整合素(integrin,ITG)α_3及ITGβ_1mRNA表达水平的影响。方法 C57BL/6小鼠右前肢腋窝皮下接种Lewis细胞建立肺癌小鼠模型,随机分为模型组、CTX组、益肺逐积方组、联合用药组,每组10只,造模次日益肺逐积方组和模型组分别给予益肺逐积方(14.95g/kg)和等体积生理盐水灌胃,1次/d;CTX组在造模后72h和10d腹腔注射CTX(40mg/kg);联合用药组在CTX组基础上灌胃益肺逐积方。给药期间观察小鼠状态及成瘤情况,给药14d后测量体重、瘤体积计算抑瘤率;常规石蜡切片检测肿瘤组织病理变化;荧光定量PCR检测肿瘤组织CD151、ITGα_3及ITGβ_1mRNA表达。结果与模型组比较,益肺逐积方组与联合用药组肿瘤体积减小(P<0.05);抑瘤率由高到低依次为:联合用药组、益肺逐积方组、CTX组。病理学检测结果显示肿瘤组织呈实片状排列并向骨骼肌浸润,肿瘤细胞沿细支气管壁及肺泡壁弥漫性生长。与模型组比较,益肺逐积方组、CTX组及联合用药组CD151mRNA表达降低(P<0.01,P<0.05),联合用药组ITGα_3mRNA表达量降低(P<0.05)。与CTX组比较,益肺逐积方组ITGβ_1mRNA的表达量降低(P<0.05)。结论益肺逐积方与CTX联合用药可更有效抑制Lewis肺癌小鼠肿瘤生长,益肺逐积方可通过下调肿瘤组织CD151和ITGα_3及ITGβ_1mRNA表达水平,达到治疗肺癌的功效。 Objective To explore the effect of Yifei Zhuji Formula(YFZJF)combined with Cyclophosphamide(CTX)on CD151 and integrin(ITG)α3 and ITGβ1 mRNA expression in tumor tissue of mice with Lewis lung cancer.Methods After the mice model of lung cancer was established by subcutaneous inoculation of Lewis cells in the right forelimb,the C57 BL/6 mice were randomly divided into 4 groups,i.e.,model group,CTX group,YFZJF group and combination group,10 in each group.The mice in YFZJF group and model group were given YFZJF(14.95 g/kg)or normal saline respectively by gavage once a day.CTX group mice received CTX(40 mg/kg)at 72 h and the 10 th day after inoculation by intraperitoneal injection.Combination group were given CTX combined with YFZJF(14.95 g/kg).The status of mice and tumor formation was observed.The mice were sacrificed after treatment for 14 days,the body mass and tumor volume were measured as well as the tumor inhibition rate was calculated.Tumor tissue pathological changes were observed by routine paraffin sectiontechnique.The expression of CD151,ITGα3 and ITGβ1 mRNA were determined by Real-time PCR.ResultsCompared with model group,the tumor volume was decreased significantly in YFZJF group and combination group(P〈0.05),and the inhibitory rate order from high to low were combination group,YFZJF group,CTX group.Pathology results showed that tumor tissue arranged in flake shape and infiltrated into skeletal muscle,the tumor cells appeared the diffuse growth along the wall of bronchioles and alveolar.The expressions of CD151 mRNA in YFZJF,CTX and combination groups were decreased significantly compared with model group(P〈0.01,P〈0.05),and ITGα3 mRNA in combination group was also decreased(P〈0.05).Compared with CTX group,the expression of ITGβ1 mRNA in YFZJF group was decreased(P〈0.05).Conclusion YFZJF combined with CTX treatments can suppress the growth of Lewis lung cancer effectively,and the effect of YFZJF may be involved in down-regulating the expression of CD151,ITGα3 and ITGβ1 mRNA in tumor tissue.
