摘要
目的:研究龙丹腰痹宁对腰椎间盘突出症模型大鼠血细胞及血清TNF-α、IL-1β、β-NGF细胞因子表达的影响。方法:采取大鼠自体髓核移植法建立腰椎间盘突出症模型。将实验大鼠分为8组,分别为空白组、假手术组、龙丹腰痹宁低、中、高剂量组、中药对照组、西药对照组。给药21 d后处死大鼠,断头取血测血细胞及其分类,ELISA法测定血清细胞因子TNF-α、IL-1β、β-NGF表达情况。结果:假手术组和模型组大鼠白细胞总数明显高于空白组(P<0.05);龙丹腰痹宁低、中、高剂量组、中药对照组、西药对照组白细胞总数均明显低于模型组(P<0.05)。模型组TNF-α、IL-1β明显高于空白组与假手术组(P<0.05);龙丹腰痹宁低、中、高剂量组、中药对照组、西药对照组TNF-α、IL-1β均明显低于模型组(P<0.05)。结论:龙丹腰痹宁可降低腰椎间盘突出症模型大鼠白细胞总数并下调血清中TNF-α、IL-1β表达,这可能是龙丹腰痹宁发挥抗炎镇痛作用的主要机制。
Objectives: To study the effects of Longdan Yaobi'ning Pellets on blood cells and serum cytokine expressions of TNF-α, IL-1β and [5-NGF in Lumbar intervertebral disc herniation rats. Methods: A rat model of lumbar intervertebral disc herniation was established by transplanting autologous nucleus pulposus. The rats were randomly classified into following 8 groups: blank group, sham group, model group, Longdanyaobining Pellets (low dose) group, Longdanyaobining Pellets (medium dose) group, Longdanyaobining Pellets (high dose) group, TCM control group, western medicine control group. 21 days after modeling all rats were executed, the blood cells and classification were tested by decapitation, the serum expressions of TNF-α, IL-1βand β-NGF were tested by enzyme linked immuno sorbent (ELISA). Results: The sham group and model group showed higher white blood cell counts than the blank group(P〈0.05); The all treat groups (Longdanyaobining Pellets low, medium and high dose groups, TCM and western control group showed lower white blood cell counts than the model group significantly (P〈0.05). Compared with blank group and sham group, model group has shown remarkable increase of TNF-α, IL-1β(P〈0.05) but no significant changes in expression of ^-NGF. Compared with model group, all treat groups(Longdanyaobining Pellets low, medium and high dose groups, TCM and western control group) has shown remarkable decrease of TNF-α, IL-1β(P〈0.05). Conclusions: Longdan Yaobi'ning Pellets can reduce the white blood cell counts and decrease the expression of TNF-α and IL-1β in Lumbar intervertebral disc herniation rats, which may be the main mechanism for Longdan Yaobi^ning Pellets to play anti-intlammatory and analgesic effects.
