摘要
目的 :探讨不同剂量的环孢素A(CsA)对大鼠慢性肾毒性模型转化生长因子 β1 (TGF β1 )、肾素mRNA表达的影响。方法 :40只雄性SD大鼠 ,随机分为 5组 ,每组 8只。A组为橄榄油对照组 ;B组CsA 1 0mg/ (kg·d)组 ;C组为CsA 2 0mg/ (kg·d)组 ;D组CsA 30mg/ (kg·d) ;E组普通饮食加橄榄油对照组。除E组外 ,其余各组大鼠给予低盐饮食。采用逆转录多聚酶链反应 (RT PCR)的方法 ,研究CsA对大鼠肾内TGF β1 、肾素mRNA的表达影响。结果 :与A组相比 ,给予CsA 1 0mg/ (kg·d)组大鼠肾内TGF β1 mRNA、肾素mRNA表达轻度增加 ,但差异无统计学意义 (P >0 .0 5) ;而给予CsA 2 0mg/ (kg·d)组大鼠肾内TGF β1 mRNA、肾素mRNA表达明显增加 (P <0 .0 5) ,CsA 30mg/ (kg·d)组增加更加明显 (P <0 .0 1 )。 结论 :CsA的慢性肾毒性与肾内TGF β1 mRNA。
Aim: To explore the effects of different dosage of cyclosporine on chronic cyclosporine induced nephrotoxicity in a rat model. Methods:Forty adult male SD rats were randomly allocated into 5 groups.Group A received olive oil as control. Group B,C,and D received CsA 10 mg/(kg·d),20 mg/(kg·d),and 30 mg/(kg·d) respectively by gavage. The rats of group A,B,C and D received low salt diet. Group E received normal salt diet and olive oil as control. The expression of mRNA for intrarenal transforming growth factor β 1 (TGF β 1) and renin mRNA were detected by reverse transcription polymerase chain reaction (RT PCR). Results: Expressions of TGF β 1 and renin mRNA were slightly higher in the rats given CsA 10 mg/(kg·d) than those in Group A, but the difference did not achieve statistical significance( P >0.05). However, they were significantly increased in the rats both given CsA 20 mg/(kg·d)( P <0.05) and given CsA 30 mg/(kg·d)( P <0.01). Conclusions: Chronic cyclosporine induced nephropathy may be related to TGF β 1 and renin mRNA up regulation. The severity of the nephropathy is related to the dosage of CsA.
