摘要
目的探讨注射用左旋泮托拉唑钠对大鼠单纯反流性食管炎的影响,为左旋泮托拉唑钠的临床开发提供依据。方法将70只大鼠随机分为7组,即模型对照组,正常对照组,左旋泮托拉唑钠低、中、高剂量组(0.9、1.8、3.6 mg/kg),右旋泮托拉唑钠组(3.6 mg/kg),泮托拉唑钠组(3.6 mg/kg)。各组尾静脉注射给予相应药物溶液,采用幽门结扎手术法造成大鼠单纯性反流性食管炎模型,检测各组食管炎指数、食管炎抑制率、胃液p H值、胃蛋白酶活性并进行比较。结果左旋泮托拉唑钠中剂量组、左旋泮托拉唑钠高剂量组、泮托拉唑钠组食管炎指数、胃蛋白酶活力低于模型对照组(P<0.05),右旋泮托拉唑钠组高于左旋泮托拉唑钠高剂量组(P<0.05);左旋泮托拉唑钠中剂量组、左旋泮托拉唑钠高剂量组、泮托拉唑钠组损伤抑制率、p H值高于左旋泮托拉唑钠低剂量组(P<0.05),右旋泮托拉唑钠组低于左旋泮托拉唑钠高剂量组(P<0.05)。结论左旋泮托拉唑钠对大鼠单纯反流性食管炎有显著的抑制作用,具有进一步临床开发价值。
Objective To explore the effect of levopantoprazole sodium on rat simple reflux esophagitis,and to provide a basis for the clinical development of levopantoprazole sodium. Methods A total of 70 rats were randomly divided into7 groups: model control group,normal control group,the levopantoprazole sodium low,medium and high group( 0. 9,1. 8,3. 6 mg/kg),the dextropantorazole sodium group( 3. 6 mg/kg),pantorazole sodium group( 3. 6 mg/kg). Each group received intravenous injection solution in tail,and simple reflux esophagitis model was established by pylorus ligation in rats. Indexes of esophagitis,esophagitis inhibition rate,p H value of gastric juice,pepsin activity were detected. Results The esophagitis index and pepsin activity in the middle dose group,the high dose group and the high dose group were lower than those in the model control group( P〈0. 05),and the dextropantoprazole sodium group was higher than that in the levopantoprazole group( P〈0. 05). The damage inhibition rate and p H value of levopantoprazole medium dose group,levopantoprazole high dose group and pantoprazole sodium group were higher than those of low dose group( P〈0. 05),and that of dextropantoprazole sodium group was lower than that of levopantoprazole sodium high dose group( P〈0. 05). Conclusion There is a significant inhibitory effect of levopantoprazole sodium on the simple reflux esophagitis in rats,which is of value for further clinical application development.
