旱半夏不同外植体的愈伤组织诱导 被引量:3

The Callus Induction from Different Explants of Pinellia ternata

摘要 本研究以四川广安地区野生旱半夏为材料,其叶片为外植体,探索其消毒条件,获得无菌苗。对旱半夏的外植体的选择部位2,4-D和KT两种激素浓度对愈伤组织诱导的影响进行研究。结果显示:选择四川广安地区5月份野生旱半夏嫩叶为外植体,70%酒精50 s,2%次氯酸钠处理12 min是最佳的灭菌条件;以MS为基本培养基,添加低浓度的激素2,4-D和KT利于旱半夏愈伤的形成,其中0.2 mg/L 2,4-D+1.0 mg/L KT叶的愈伤诱导率达98.3%,0.2 mg/L 2,4-D+0.5mg/L KT叶柄的愈伤诱导率达91.8%,0.5 mg/L 2,4-D+0.5 mg/L KT珠芽的愈伤诱导率达92.8%,且长势旺盛。对旱半夏的叶、叶柄、珠芽三种外植体进行比较,发现其叶片更利于愈伤的诱导,不仅其愈伤诱导率高且再生苗生长好。在此研究的基础上优化培养条件,为旱半夏的再生、生根培养、提高繁殖系数等生理研究提供了理论依据。 Using wild Pinell ia ternata in Guang'an area of Sichuan province as test materials and its leaves as explants,the study explored their disinfection conditions and obtained aseptic seedling.The selection part of explants,and the effects of 2,4-D and KT hormone concentrations on callus induction were studied.The results showed that selecting the tender leaves of wild Pinellia ternate from Guang'an,Sichuan in May as explants,the treatment with 70%alcohol for 50 s and 2% sodium hypochlorite for 12 min was the best sterilization condition.With MS as the basic culture medium,the addition of low concentration of hormones 2,4-D and KT was conducive to the formation of Pinellia ternata callus.The callus induction rate of leaves was 98.3% at 0.2 mg/L 2,4-D with 1.0 mg/L KT,and that of petiole was 91.8% at 0.2 mg/L 2,4-D with 0.5 mg/L KT.The callus induction rate of bulbil was 92.8% at 0.5 mg/L2,4-D with 0.5 mg/L KT,meanwhile,they were growing strong.It was found that the leaves were more favorable to the induction of callus,which was not only shown in the higher rate of callus,but also the growth of the regeneration seedlings.On the basis of this study,the optimized training conditions might provide theoretical basis for physiological studies such as regeneration,rooting culture and improving reproduction coefficient of Pinellia ternata.
