Transcriptional Analysis of Two Different Kinds of Gastrodia elata Seeds

摘要 目的:对两种天麻种子进行转录组分析,在转录组水平研究两种天麻种子次级代谢产物含量与相关酶活性的关系,从而对其进行品质评价。方法:采用Illumina Hiseq 2500对乌天麻种子及乌红杂交天麻种子进行转录组测序分析,通过与基因数据库比对、分析、注释来发现差异表达基因。结果:乌天麻种子及乌红杂交天麻种子分别获得了24 892 730、29 712 119条Clean Reads,Trinity组装后得到383 242条Unigenes,通过六大基因数据库比对、分析、注释,最终获得184 706个有注释信息的Unigenes。与乌天麻种子相比,杂交后的天麻种子有178个基因表达量上调,527个基因表达量下调,合计705个差异表达基因。其中共有10个关键基因被KEGG数据库苯丙烷类合成途径注释,表达量下调的差异表达基因有PAL、β-G、POD和aroX,表达量上调的有NADP+。结论:本研究获得了乌天麻种子和乌红杂交天麻种子的苯丙烷类产物合成代谢通路图,并注释了相关基因序列,最终确定乌红杂交天麻种子品质最佳。 Objective:To analyze two kinds of Gastrodia elate seeds by transcriptionapt method, and to study the relationship between the content of secondary metabolites and the activities of related enzymes in two different kinds of Gastrodia elata seeds and evaluate their quality basised on transcriptome analysis. Methods:Illumina Hiseq :2500 was used to detect the seeds of Wu Gastrodia elata and Wuhong hybrid Gastrodia elata in transcriptome sequencing analysis, differentially expressed genes were identified by comparing, analyzing and annotating with gene database. Resuhs:24 892 730 and 29 712 119 Clean Reads were obtained respectively in the seeds of Wu Gastrodia elata and Wuhong hybrid Gastrodia elata, and 383 242 Unigenes were obtained after Trinity assembly. 184 706 annota- ted Unigenes were obtained finally by comparing, analysing and annotating through six Gene Database. Comparing with the seeds of Wu Gastrodia elata, the seeds of Wuhong hybrid Gastrodia elata has total 705 differently expressed genes,including 178 genes expression in- cresed and 527 genes expression decreased, and ten key genes differential expression data were annotated by KEGG database into phe- nylpropanoid metabolic pathway. The expression of PAL,β-G,POD,aroX decreased, and the expression of NADP+ increased. Conclusion : This study has obtained a complete metabolic pathway map of the product of two different kinds of Gastrodia elata seeds, and annotate a large number of related gene sequences, and finally determine the seeds of Wuhong hybrid Gastrodia elata with higher quality.
