摘要
为蛤蚧酒中多糖含量测定方法的建立提供试验依据,采用苯酚-硫酸法测定蛤蚧酒中多糖含量。考虑到酒中成分复杂,可能对多糖含量测定产生影响,因此用沉淀剂醋酸铅除去干扰物质,比较使用沉淀剂和不使用沉淀剂对蛤蚧酒多糖含量测定的影响。苯酚-硫酸法建立的葡萄糖标准曲线为A=8.388 89C+0.001 51,相关系数r^2=0.998 82,线性范围为0.00~0.10 mg/mL,平均回收率为99.22%,方法检出限为0.89 mg/L,方法相对标准偏差为2.46%。使用沉淀剂前后样品的吸光度差异不显著(p>0.05),说明酒中其他成分对于蛤蚧酒中多糖含量的测定没有影响,使用苯酚-硫酸法测定蛤蚧酒中多糖含量时不需要使用沉淀剂除去其他成分。综上,苯酚-硫酸法可作为蛤蚧酒中多糖含量测定的方法;其他成分对测定结果的干扰可忽略。
To provide the experimental basis for the determination of polysaccharide content in the alcoholic drink of Gekko gecko Linnaeus, the content of polysaccharide in the alcoholic drink of Gekko gecko Linnaeus was determined by phenol-sulfuric acid colorimetry method. Take into account the impact of interference in the wine, with the precipitant lead acetate to remove the interference material, compare the use of precipitants and do not use precipitants on the alcoholic drink of Gekko geckoLinnaeus polysaccharide content determination. The standard curve of glucose established by phenol-sulfuric acid colorimetry method was A=8.388 89 C+0.001 51, the correlation coefficient r^2 was 0.998 82, the linear range of the standard curve was 0.00-0.10 mg/mL, the average recovery was 99.22%, the detection limit was 0.89 mg/L, and the relative standard deviation of the method was 2.46%. The absorbance value difference of the sample before and after precipitation treatment was not significant(P〉0.05), indicating that other components in the alcoholic drink had no effect on the determination of the polysaccharide content in the alcoholic drink of Gekko gecko, and the use of the phenol-sulfuric acid method did not require the use of a precipitant when determining the polysaccharide content in the alcoholic drink of Gekko gecko to remove other components. In summary, the phenol-sulfuric acid method could be used as a method for the determination of the polysaccharide content in the alcoholic drink of Gekko gecko, and the interference of other components with the assay results could be ignored.
