高效液相色谱法分析蔗糖脂肪酸酯商品 被引量:2

Analysis of Commercial Sucrose Esters by High Performance Liquid Chromatography

摘要 采用高效液相色谱-质谱联用技术(HPLC-MS)和高效液相色谱-蒸发光散射检测法(HPLC-ELSD)对蔗糖脂肪酸酯商品进行了定性和定量分析。考察了HPLC条件:Symmetry C18(4.6 mm×75 mm,3.5μm)为色谱柱,[V(甲醇)︰V(四氢呋喃)=80︰20]-水二元溶剂为流动相,梯度洗脱,流速为1.0 mL/min,柱温为35℃,分析时间为23 min,以及ELSD条件:雾化气为空气,压力为2.0 bar,漂移管温度为40℃。研究结果表明:在该条件下,HPLC-ELSD能够有效分离与测定蔗糖酯商品中的蔗糖、蔗糖单酯、蔗糖二酯及蔗糖三酯,蔗糖单酯的线性范围为0.3~2.1mg/mL(R^2=0.9992),平均回收率为102.1%,RSD为1.2%。该方法是一种简单、快速、准确可靠的蔗糖酯检测方法,可用于蔗糖酯商品的组成分析和品质监控。 The commercial sucrose fatty acid esters were qualitatively and quantitatively analyzed by high performance liquid chromatography coupled with electrospray ionization mass spectrometry(ESI-MS) and evaporative light scattering detector(ELSD). The samples were separated by a Symmetry C18(4.6 mm×75 mm, 3.5 μm) reversed phase column using V(methanol)︰V(tetrahydrofuran)=80︰20 and water as the mobile phase under a gradient condition with a flow rate of 1.0 mL/min within 23 min. The column temperature was set at 35 ℃. The eluents were determined by ELSD using air as atomizing gas of 2.0 bar. The temperature in drift tube was set at 40 ℃. Under this procedure, the sucrose, sucrose monoester, sucrose diester, and sucrose triester were completely separated and determined. A linear calibration curve of sucrose monoester was established among 0.3~2.1 mg/mL with a regression correlation coefficient of 0.9992. The average spike recovery was 102.1 %, and the relative standard deviation(RSD) was 1.2 %. The described method is useful for the component analysis and the quality monitoring of commercial sucrose esters, being simple to operate and rapid to output results with good accuracy and reliability.
