沙田柚乙烯应答因子的鉴定和生物信息学分析

摘要 利用高通量测序对沙田柚自交和异交花柱进行转录组测序,通过差异分析得到了沙田柚乙烯应答因子基因序列。采用生物信息学方法,对其编码的蛋白质从序列特征、理化性质、跨膜结构域、高级结构以及功能域等方面进行了预测和分析。该基因全长为1013 bp(Gen Bank登录号为MG905213),开放阅读框(ORF)全长498 bp,编码的蛋白质含166个氨基酸。编码蛋白质的相对分子量为18.45 k Da,理论等电点为7.90。该蛋白质具有一个与AP2 superfamily蛋白相同的保守结构域,为亲水性非分泌不稳定蛋白,不属于跨膜运动,共有24个可能的磷酸化位点。氨基酸序列分析表明,该基因编码的蛋白质与芸香科的甜橙(Citrus sinensis,XP-006467696.1)和克莱门柚(Citrus clementina,XP-006449381.1)的AP2蛋白的同源性分别为100%和98%。系统进化树表明,沙田柚的乙烯应答转录因子与甜橙和克莱门柚亲缘关系很近,属于同一进化分支。 The transcriptomes of the self-pollinated style and cross-pollinated style of Citrus grandis var. shatinyu Hort were researched by high-throughput sequencing,and the ethylene response factor gene sequence was obtained from this plant through difference analysis. The sequence characteristics,physicochemical properties,transmembrane domains,advanced structures and functional domains of the proteins encoded by this gene were analyzed and predicted by using bioinformatic method. The results showed that the full length of the gene was 1013 bp(Gen Bank login number was MG905213),the open reading frame(ORF)was 498 bp in full length,encoding 166 amino acids with deduced molecular weight of 18. 45 k Da,and theoretical p I value was7.90. This protein had a conserved domain,which was the same as that of AP2 superfamily protein; it was a hydrophilic non-secretory unstable protein with 24 possible phosphorylation sites,and was not involved in transmembrane movement. The homology analysis of amino acid sequence indicated that the protein encoded by this gene shared high homology with the AP2 protein of Citrus sinensis,XP-006467696.1(100%) and Citrus clementina,XP-006449381.1(98%). The phylogenetic tree indicated that the ethylene response factor in C. grandis var. shatinyu had very close kinship with that in C. sinensis and C. clementina,and they belonged to the same evolutionary branch.
