摘要
目前防控柑橘黄龙病的措施主要包括培育无病毒苗、防治木虱、铲除病树等。而多数防控措施的开展均需灵敏、可靠的早期诊断技术配合。综述了柑橘黄龙病的田间诊断方法以及基于电镜、血清学、高光谱、淀粉显色、常规PCR、巢式PCR、LAMP PCR、定量PCR、数字PCR的检测方法 ,并阐述了各自的优缺点。其中,定量PCR是目前较成熟和具有较高灵敏度的检测技术,可配合其他低成本检测方法对未显症、假阴性等样品进行早期检测;可实现单分子DNA绝对定量分析,数字PCR检测技术在柑橘黄龙病检测中也具有较好的应用前景。
At present, major measures against citrus Huanglongbing(HLB) includes planting of disease-free nursery stock,controlling psyllid vector by spraying pesticides and removal of infected trees. Most of these controlling mesures need sensitive and reliable early diagnosis technologies. This paper reviewed field diagnosis and detection methods for citrus Huanglongbing, associated with electron microscope, serology, hyperspectra, iodine-starch color development, conventional PCR, nested-PCR, LAMP PCR, real-time quantitative PCR and digital PCR, and evaluated their advantages and drawbacks. Among them, real-time quantitative PCR was relatively mature and sensitive detection technology currently,which can be used with other low-cost detection methods to conduct early detection for asymptomatic and false-negative samples. Besides, the digital PCR, which could realize absolute quantification of single molecule DNA, also had a good application prospect in the detection of citrus Huanglongbing.
