仙鹤草水提液对胰腺癌细胞BXPC-3和PANC-1增殖的抑制作用研究 被引量:12

Inhibitory Effects of Agrimoniae Herba Extracting Solution on Pancreatic Cancer Cell BXPC-3 and PANC-1 Proliferation

摘要 目的研究仙鹤草水提液对胰腺癌细胞BXPC-3和PANC-1增殖的抑制作用。方法 SRB法检测不同浓度仙鹤草水提液对胰腺癌细胞BXPC-3(仙鹤草水提液浓度:0.15,0.3,0.6,1.2,2.4,4.8 mg·m L-1)和PANC-1(仙鹤草水提液浓度:0.125,0.25,0.5,1.0,2.0,4.0 mg·m L-1)分别作用24,48,72 h后的增殖抑制率;流式细胞仪检测0.5,1,2 mg·m L-1仙鹤草水提液对胰腺癌细胞BXPC-3周期分布的影响;Annexin V-FITC/PI双染法检测1,2,4,8 mg·m L-1仙鹤草水提液对BXPC-3细胞凋亡的影响。结果仙鹤草水提液可显著抑制BXPC-3和PANC-1细胞的增殖,并呈时间和浓度依赖性;仙鹤草水提液可显著增加BXPC-3细胞停滞在G1期的百分率,促进BXPC-3细胞的早期凋亡和晚期凋亡。结论仙鹤草水提液可通过或部分通过诱导胰腺癌细胞BXPC-3和PANC-1的凋亡,抑制胰腺癌细胞的增殖,发挥抗胰腺癌作用。 OBJECTIVE To investigate the inhibitory effects of Agrimoniae Herba extracting solution on different pancreatic cancer cell(BXPC-3 and PANC^-1) proliferation. METHODS The effects of different concentration of Agrimoniae Herba extracting solution on the inhibitory effect of BXPC-3(Agrimonia Herb extracting solution: 0.15, 0.3, 0.6, 1.2, 2.4, 4.8 mg·m L^-1) and PANC^-1(Agrimoniae Herba extracting solution: 0.125, 0.25, 0.5, 1.0, 2.0, 4.0 mg·m L^-1) proliferation for 24, 48, 72 h were detected by SRB method. The effect of Agrimoniae Herba extracting solution(0.5, 1, 2 mg·m L^-1)(BXPC-3) on cell cycle was detected by flow cytometry. The effect of Agrimoniae Herba extracting solution(1, 2, 4, 8 mg·m L^-1)(BXPC-3) on cell apoptosis was detected by Annexin V-FITC/PI staining. RESULTS The proliferation of BXPC-3 cells were remarkably inhibited by Agrimoniae Herba extracting solution at a time and concentration dependent manner. The rate of cells in G1 were enhanced, and the cell apoptosis were promoted by Agrimoniae Herba extracting solution. CONCLUSION Agrimoniae Herba extracting solution inhibits proliferation of BXPC-3 and PANC^-1 cells via or partly via inducing cell apoptosis and plays the role of anti-pancreatic cancer.
