摘要
目的:研究人脐带间充质干细胞(HUC-MSCs)对舌鳞癌基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)表达的影响。方法:培养Cal-27细胞和经绿色荧光蛋白(GFP)标记的HUC-MSCs细胞,并建立26只裸小鼠Cal-27舌鳞癌移植瘤模型,成瘤后随机分实验组和对照组,每组13只。实验组尾静脉注射GFP标记的人脐带间充质干细胞,对照组注射等量生理盐水。免疫组织化学法检测移植瘤组织中GFP蛋白表达情况,以及MMP-2、MMP-9蛋白表达水平;实时荧光定量PCR检测MMP-2和MMP-9mRNA的相对表达量。结果:实验组肿瘤组织GFP表达阳性,对照组GFP表达阴性;实验组MMP-2和MMP-9表达量均明显低于对照组,差异均有统计学意义(P<0.05)。结论:HUC-MSCs有向舌鳞癌的归巢性,下调肿瘤组织MMP-2和MMP-9的相对表达量,提示HUC-MSCs具有抑制舌鳞癌Cal-27细胞侵袭转移的潜力。
Objective： To investigate the effect of human umbilical cord mesenchymal stem cells（ HUC-MSCs） on the expressions of matrix metalloproteinase-2（ MMP-2） and matrix metalloproteinase-9（ MMP-9） in the tongue squamous cell carcinoma. Methods： Cal-27 cells and HUC-MSCs（ marked by GFP） were cultured,and animal models of Cal-27 tongue squamous cell carcinoma transplantation tumor were established in 26 nude mice that were randomly divided into experimental group and control group,with 13 mice in each group. HUC-MSCs injection via the tail vein in experimental group,the same volume of normal saline in control group. The expression of GFP,MMP-2 and MMP-9 were detected by immunohistochemistry and the mRNA level of MMP-2 and MMP-9 was detected by real-time fluorescence quantitative PCR. Results： GFP expression was positive in experimental group and negative in control group. The expression of MMP-2 and MMP-9 in the experimental group was significantly lower than that in the control group respetively（ P〈0. 05）. Conclusion： HUC-MSCs with the characteristics of tumor homing can reduce the expression of MMP-2 and MMP-9 in tumor tissues,suggesting that HUCMSCs with the potential of inhibiting the invasion and metastasis of Cal-27 cancer cells.
