摘要
目的:研究加味小柴胡汤各提取部位的有效成分对幽门螺杆菌(Hp)的体外抑制效果。方法:对6-姜酚、盐酸小檗碱、黄芩苷、柴胡皂苷a等有效成分进行抑菌试验,琼脂扩散实验采用牛津杯的方法,按一定菌浓度混合培养基铺菌层,每孔100μl药液,培养3 d,测量抑菌圈直径;液体稀释实验采用梯度稀释法,在试管内加入1 ml含药培养基和50μl菌液,培养3d,观察浑浊情况并计算最小抑菌浓度(MIC)。结果:6-姜酚、盐酸小檗碱、黄芩苷、柴胡皂苷a等有效成分抑菌效果均优于阳性对照组(P<0.01);盐酸小檗碱组的抑菌效果优于其他3个给药组(P<0.05)。结论:确定了6-姜酚、盐酸小檗碱、黄芩苷、柴胡皂苷a等有效成分均对Hp有抑制作用,为研究加味小柴胡汤方抑制Hp的物质基础提供科学依据。
Objective: To investigate the inhibitory effect of the effective components in the different extracts of modified Xiao Chaihu decoction on Helicobacter pylori ( H. pylori) in vitro. Methods: 6-Gingerol, berberine hydroehloride, baiealin, and saikosapo- nin a were tested for bacteriestasis. Agar diffusion test was carried out by using Oxford cup method. The medium was mixed with a cer- tain concentration of bacteria with 100 μl per well and cultured for 3 days, and then the diameter of inhibition zone was measured. The liquid dilution experiment was performed with gradient dilution ,50 μl bacteria liquid and 1 ml medium containing drug were mixed and cultured for 3 days, and the turbidity was observed and the minimal inhibitory eoncentratio (MIC) was calculated. Results : The anti- bacterial effects of 6-gingerol, berberine hydroehloride, baicalin and saikosaponin a were all better than that of the positive control group ( P 〈 0.01 ), and berberine hydroehloridc group had better antibacterial effect than the other three groups ( P 〈 0.05 ). Conclusion: The results proved that 6-gingerol, berberine hydrochloride, baicalin and saikosaponin a all have inhibitory effects on H. pylori, which provide scientific evidence for the study on the material basis of modified Xiao Chaihu decoction used for inhibiting H. pylori.
