摘要
目的研究金钗石斛多糖(DNP)对秀丽隐杆线虫衰老的影响,并初步探讨其机制。方法设立金钗石斛多糖组(0.06、0.12、0.24 mg/m L)、PBS缓冲液组及空白对照组(0 mg/L),观察金钗石斛多糖对N2野生型秀丽隐杆线虫的寿命及生理指标(生殖能力、咽泵频率和身体摆动频率)的影响。通过进行线虫的氧化应激实验和热应激实验,初步探讨金钗石斛多糖延缓衰老的机制。结果在寿命实验中,金钗石斛多糖组线虫的平均寿命较空白对照组显著延长(P<0.05),PBS缓冲液组较空白组差异无统计学意义(P>0.05);在生殖能力、咽泵频率和身体摆动频率实验中,金钗石斛多糖组较空白对照组差异无统计学意义(P>0.05);在氧化应激实验和热应激实验中,金钗石斛多糖组线虫的平均生存时间较空白对照组明显延长(P<0.05)。结论金钗石斛多糖能延缓秀丽隐杆线虫衰老,其机制可能与金钗石斛多糖提高线虫的抗应激能力有关。
Objective To study the aging effect of Dendrobium nobile polysaccharides( DNP) on caenorhabditis elegans and to preliminary discussion about its mechanism. Methods DNP groups( 0. 06,0. 12,0. 24 mg/m L),PBS buffer and blank control group( 0 mg/L) were established to investigate the effects of DNP on the lifespan and physiological indexes( reproductive capacity,pumping rate and body swing frequency) of N2 wild-type caenorhabditis elegans. To perform oxidative and heat stress test of elegans,and preliminary? investigate the mechanism of DNP retard aging. Results In lifespan test,average lifespan of elegans in DNP groups were prolonged significantly compared to blank control group( P〈0. 05) and PBS buffer group was no difference compared to blank control group( P〉0. 05). In reproductive capacity,pumping rate and body swing frequency test,the reproductive capacity,pumping rate and body swing frequency of elegans in DNP groups were no difference compared to blank control group( P〉0. 05). In oxidative and heat stress test,average survival time of elegans in DNP group was prolonged significantly compared to blank control group( P〈0. 05). Conclusion DNP can retard the aging process of caenorhabditis elegans,the mechanism might involve increase of the stress resistance ability of elegans.
