摘要
目的:观察大黄?虫丸对糖尿病大鼠视网膜毛细血管中PI3K-AKT信号通路相关因子表达的影响,探讨大黄?虫丸对糖尿病视网膜病变(DR)微血管损伤的保护作用及机制。方法:100只雄性SPF级Wistar大鼠随机分成空白组,模型组,大黄?虫丸低、中、高剂量组。除空白组外各组均采用链佐脲菌素腹腔注射诱导糖尿病大鼠模型。造模成功后分别于给药6、12周后取大鼠眼球进行检测。RT-PCR定量检测PI3K、AKT mRNA在视网膜组织中的表达水平。Western blot法检测各组PI3K、AKT、Caspase-9在视网膜组织中的蛋白表达量。结果:与空白组比较,模型组视网膜中PI3K、AKT mRNA表达水平下降,PI3K、AKT蛋白相对表达量下降(P<0.05),而Caspase-9蛋白相对表达量上升(P<0.05);灌胃6、12周后,大黄?虫丸各剂量组与模型组比较,视网膜中PI3K、AKT mRNA含量均升高,PI3K、AKT蛋白相对表达量升高(P<0.05),Caspase-9蛋白相对表达量下降(P<0.05),其中,大黄?虫丸中剂量组PI3K、AKT mRNA表达最接近空白组。结论:大黄?虫丸能够上调PI3K、AKT,降低Caspase-9表达,抑制糖尿病大鼠视网膜微血管周细胞的凋亡,从而发挥其对DR微血管损伤的保护作用。
Objective: To observe the effects of Dahuang Zhechong Pill on the expression of PI3 K-AKT signaling pathway related factors in retinal capillaries of diabetic rats, to explore the protective effect and mechanism of Dahuang Zhechong Pill on diabetic retinopathy microvascular injury. Methods: A total of 100 male SPF Wistar rats were randomly divided into blank group, model group, Dahuang Zhechong Pill low dose group, middle dose group and high dose group. Except the blank group, DR rats were induced by intraperitoneal injection of streptozotocin in each group. After successful modeling, rats were examined for eyeballs of 6 weeks and 12 weeks administration respectively. PI3 K and AKT mRNA expression in retina tissue were detected by RT-PCR. The expression of PI3 K, AKT and Caspase-9 in the retina tissue were detected by Western blot. Results: Compared with the blank group, the mRNA expression levels of PI3 K and AKT in the retinal model group were decreased, the protein expression levels of PI3 K and AKT were decreased(P〈0.05), and Caspase-9 was increased(P〈0.05); after gavaging for 6 weeks and 12 weeks, compared with model group, the PI3 K and, AKT mRNA in retina of Dahuang Zhechong Pill group were increased, the protein expression levels of PI3 K and AKT were increased(P〈0.05), and Caspase-9 was decreased(P〈0.05). What's more, the expression of PI3 K and AKT mRNA in Dahuang Zhechong Pill middle dose group was the closest to the normal blank group. Conclusion: Dahuang Zhechong Pill can increase the expression of PI3 K and AKT decrease the expression of Caspase-9, and inhibit the apoptosis of retinal microvascular pericytes in DM rats, which exert its protective effect on diabetic retinopathy microvascular injury.
