摘要
目的优化复合酶酶解蜈蚣粉最佳酶解工艺。方法采用正交试验法探究酶解温度、酶解时间、pH值及单个酶酶活力对蜈蚣复合酶酶解液冻干粉对乳腺癌MCF-7细胞增殖抑制率的影响,并确定最佳酶解工艺条件。结果蜈蚣复合酶最佳酶解工艺条件为酶解温度35℃、酶解时间3 h、pH值为8.0及单个酶酶活力2000 U/g,在此工艺条件下,蜈蚣复合酶酶解液冻干粉对乳腺癌MCF-7细胞的增殖抑制率为61.67%。结论通过实验验证此工艺可行,蜈蚣复合酶酶解液冻干粉对乳腺癌MCF-7细胞的增殖抑制率较高。
Objective To optimize the enzymatic hydrolysis process of centipede powder with complex enzyme. Methods The orthogonal test was adopted to explore the effects of enzymatic hydrolysis temperature, enzymatic hydrolysis time, pH value and activity of single enzyme on the inhibition rate on the breast cancer MCF-7 cells and determine the optimum enzymatic hydrolysis conditions. Results The optimum conditions were as follows: the enzymatic hydrolysis temperature was 35 ℃, the enzymatic hydrolysis time was 3 h, the pH value was 8.0 and activity of single enzyme was 2000 U/g. Under these conditions, the lyophilized powder of the enzymatic hydrolysate showed the inhibition rate on the proliferation of the breast cancer MCF-7 cells of 61.67 %. Conclusion This enzymatic hydrolysis process is feasible, and the lyophilized powder of the enzymatic hydrolysate has a strong inhibition effect on the breast cancer MCF-7 cells.
