摘要
目的:探讨化脓性链球菌定植后IL-17A的表达及与生殖道炎症因子的关系。方法:选择健康清洁级雌性Balb/c小鼠48只,随机分为实验组(n=24)和对照组(n=24)。实验组小鼠建立A群链球菌阴道炎小鼠模型;对照组小鼠经生理盐水处理。收集两组小鼠阴道分泌物涂片,于接种后7d、14d取小鼠阴道组织进行PAS染色和HE染色,并作组织匀浆处理及ELISA测定。比较两组IL-17A mRNA、蛋白相对表达量及IL-17A水平与生殖道炎症因子的相关性。结果:接种后14d,小鼠外阴明显红肿,阴道分泌物涂片HE染色发现实验组小鼠在上皮组织中有大量中性粒细胞存在。接种后14d,实验组在IL-17A mRNA相对表达量、IL-17A蛋白相对表达量方面显著高于对照组(P<0.05)。接种后7d、14d,实验组在TNF-α、IL-1β、IL-22、IL-6方面显著高于对照组(P<0.05),差异具有统计学意义;相比于接种后7d,接种后14d在TNF-α、IL-1β、IL-22、IL-6方面差异显著(P<0.05,具有统计学意义)。接种后7d、14d,实验组小鼠血清IL-17A水平显著高于对照组(P<0.05),差异具有统计学意义。IL-17A与IL-1β呈正相关(P<0.05),差异具有统计学意义;与TNF-α、IL-22、IL-6呈负相关(P<0.05),差异无统计学意义。结论:IL-17A表达上调可能在A群链球菌致病中起到重要的免疫调节作用。
Objective: To investigate the relationship between the expression of IL-17 A and the inflammatory factors in the genital tract after the colonization of Streptococcus pyogenes. Methods: 48 healthy and clean grade female Balb/c mice were selected and randomly divided into experimental group (n = 24) and control group (n =24). Mice in experimental group were established group A Streptococcus vaginitis model,while control group mice were treated with physiological saline. The vaginal smears of two groups of mice were collected,and the tissues of mice were taken 7 d and 14 d after inoculation,PAS staining and HE staining were performed,and the tissue homogenates were processed and ELISA was determined. The correlation of IL-17 A,mRNA,protein relative expression and IL-17 A levels with reproductive tract inflammatory factors in the two groups were compared. Results: After inoculation with 14 d,the vulva of mice was obviously red and swollen,and HE staining of vaginal secretions showed that there were large numbers of neutrophils in the epithelial tissue of the experimental group. After inoculation with14 d,the relative expression of IL-17 A and mRNA and the relative expression of IL-17A protein in the experimental group were significantly higher than those in the control group (P〈 0. 05). After inoculation with 7 d,14 d,experimental group was significantly higher than that in TNF-α and IL-1β,IL-22,IL-6 control group (P〈 0. 05); Compared to 7 d after inoculation,14 d after inoculation in TNF-αand IL-1β,IL-22 and IL-6 had significant difference (P〈 0. 05). The serum IL-17A level of the experimental group was significantly higher than that of the control group (P〈 0. 05). IL-17 A was positively correlated with IL-1β (P〈 0. 05),and negatively correlated with TNF-α,IL-22 and IL-6 (P〈 0. 05). Conclusons: Up regulation of IL-17A expression may play an important immunomodulatory role in the pathogenesis of group A streptococci.
