利肺片对放射性肺炎小鼠肺部炎性因子及氧化应激指标的影响 被引量:6

摘要 目的探讨利肺片对放射性肺炎小鼠肺部炎性因子及氧化应激指标的影响。方法将60只清洁级C57BL/6雌鼠随机分为空白组、模型组、地塞米松组、利肺片高剂量组、利肺片中剂量组、利肺片低剂量组,每组10只。每天对除空白组外小鼠进行3 Gy胸部照射,照射结束后1 h,地塞米松组灌胃地塞米松2. 46 mg/kg,利肺片高、中、低剂量组分别灌胃利肺片1. 1 g/kg、0. 55 g/kg、0. 252 g/kg,空白组及模型组灌胃等量生理盐水。实验30 d后处死各组小鼠,检测左侧肺组织匀浆中氧化应激指标超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)水平和炎性因子指标白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-12(IL-12)水平;取右侧肺组织分别行HE染色观察其病理变化、Masson染色观察其胶原沉淀情况,并分别进行肺泡炎及胶原沉积评分。结果利肺片各剂量组和地塞米松组小鼠肺组织匀浆中GSH-Px、SOD、CAT水平均明显高于模型组(P均<0. 05),而IL-6、TNF-α、IL-12水平均明显低于模型组(P均<0. 05)。HE及Masson染色显示,空白组无肺泡炎改变及胶原沉积,模型组多为中级以上肺泡炎及胶原沉积,而利肺片各剂量组和地塞米松组主要出现轻度的肺泡炎改变及胶原沉积,且利肺片各剂量组和地塞米松组肺泡炎及胶原沉积评分均明显低于模型组(P均<0. 05)。结论利肺片可有效减轻放射性肺炎小鼠肺部炎症及局部氧化应激反应,缓解肺纤维化。 Objective It is to investigate the effects of Lifeipian on inflammatory factors and oxidative stress in lung of radiation pneumonia mice.Methods Sixty clean grade C57BL/6 female rats were randomly divided into blank group, model group, dexamethasone group, and Lifeipian high, middle, low dose group, with 10 rats in each group. The mice were exposed to 3 Gy chest irradiation except the control group daily. One hour after irradiation, the mice were treated with intragastric administration of dexamethasone 2.46 mg/kg in the dexamethasone group, and the mice were treated with Lifeipian of 1.1 g/kg, 0.55 g/kg and 0.252 g/kg for intragastric administration in the Lifeipian high, medium and low dose groups respectively. The blank group and the model group were given the same amount of normal saline for intragastric administration. After 30 days, all mice were killed. Pulmonary tissue homogenate was prepared from some lung tissues and the levels of oxidative stress indexes such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) and inflammatory factors such as interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), interleukin-12 (IL-12) were measured; the other lung tissues were stained to observe the pathological changes by HE staining, and observe the collagen deposition by Masson staining, and scores of alveolitis and collagen deposition were recorded.Results The levels of GSH-Px, SOD and CAT in lung homogenate of the Lifeipian high, middle, low dose group and the dexamethasone group were significantly higher than those of the model group (all P 〈0.05), but the levels of IL-6, TNF-α and IL-12 were significantly lower than those in the model group (all P 〈0.05). HE and Masson staining showed that there was no alveolar inflammation and collagen deposition in the blank group, but more middle grade alveolitis and collagen deposition in the model group, however, there were mild alveolar inflammation and collagen deposition in the Lifeipian groups and the dexamethasone group, and the scores of alveolitis and collagen deposition in the Lifeipian each dose group and the dexamethasone group were significantly lower than those in the model group (all P 〈0.05).Conclusion Lifeipian has a good therapeutic effect on radiation pneumonitis mice. It can effectively improve the lung inflammation and oxidative stress disorders.
