摘要
为了解猪流行腹泻病毒(PEDV)的遗传变异,试验对来自安徽省不同地区8个猪场病料样品,利用RT-PCR方法,扩增PEDV ORF3基因,测序并进行序列分析。结果显示,8份样品均扩增出包含ORF3基因全长(675 bp)的目的片段,核苷酸序列同源性为98.5%~100%;它们与疫苗株(attenuated DR13、CH-BJ-2011 truncated)及CV777株和LZC株的同源性较低(95.1%~98.1%),与2011年后的中国及美国分离株存在较高同源性(98.2%~100%)。提示8个PEDV分离株均为强毒株。
To study the genetic variation of porcine epidemic diarrhea virus (PEDV) , the ORF3 genes of 8 PEDV field strains were cloned, sequenced and analyzed, which were isolated from different regions of Anhui Province from 2013 to 2014. The results showed that the ORF3 genes of 8 PEDV field strains were all 675 bp, and had 98.5%~100% nucleotide identity. The ORF3 genes of the 8 PEDV field strains had the lower homology (95.1%~98.1%) with attenuated DR13, CH-BJ-2011 truncated, CV777 and LZC strains, had the higher homology (98.2%~100%) with the reference PEDV strains isolated after 2011 from China and America. The re-sults suggested that the 8 PEDV field strains were all virulent strains.
