复方地黄颗粒对阴虚动风证帕金森病异动症模型大鼠神经行为学的影响 被引量:9

摘要 目的观察复方地黄颗粒对阴虚动风证帕金森病(PD)异动症模型大鼠神经行为学的影响。方法采用6-羟基多巴胺(6-OHDA)脑立体定向注射术建立大鼠PD模型,向PD大鼠腹腔注射左旋多巴(50mg/kg)/苄丝肼(12.5mg/kg),每日2次,连续2周,构建阴虚动风证PD异动症(LID)模型。将LID模型大鼠随机分为模型组和复方地黄颗粒组,每组10只,复方地黄颗粒组给予中药复方地黄颗粒(14g/kg,2ml)灌胃,模型组给予等体积生理盐水灌胃,每日1次,连续用药6周;另设正常组及假手术组,每组10只。在给药第4、6周分别观察PD大鼠LID模型异常不自主动作(AIM)评分、旋转起动时间、旋转持续时间、旋转圈数、剂峰旋转圈数、APO诱导后旋转圈数等神经行为学的变化。结果复方地黄颗粒干预后,LID大鼠AIM评分显著降低,旋转起动时间延长,旋转持续时间减低,旋转圈数、剂峰旋转圈数及APO诱导后旋转圈数减少,与模型组比较差异有统计学意义(P<0.01)。结论复方地黄颗粒可改善帕金森病LID模型大鼠的神经行为学表现,具有抗LID作用。 Objective To explore the effect of compound Rehmannia granule on neurobehavioral of Levodopa-induced dyskinesias (LID) using Parkinson's disease (PD) rat models caused by Yin deficiency and dynamic wind syndrome. Methods The PD model of rats was established by stereotactic injection to the brain with 6-hydroxydopamine. To establish the LID model from the PD rats, PD rats was further intraperitoneally injected with levodopa (50mg/kg)/Benserazide (12.5mg/kg) twice a day for 2 weeks. Further, the LID model rats were administrated with compound Rehmannia granule using oral gavage, at a dose of 2ml of 14g/kg, daily for 6 weeks (compound Rehmannia granule group, CRG group). The LID rats that received saline buffer (model group), normal rats without any treatment (normal group), and rats that received the sham operation (sham operation group), were served as control. A total of ten rats were assigned to each group. After treating rats with compound Rehmannia granule for 4 weeks and 6 weeks, the changes of neurological behaviors (AIM score, rotation start time, rotation duration, rotation circle number, peak rotation circle number, APO induced rotation number) were examined in Parkinson's disease LID model rats. Results After the intervention of the compound Rehmannia granules, the AIM score of LID rats was significantly reduced, the rotation start time was prolonged, the rotation duration decreased, the number of rotation circles decreased, the number of peak-dose rotation circles decreased, the number of rotation circles decreased after APO induction. Compared to the model group, the neurological behaviors were statistically significant (P〈O.O1). Conclusion The compound Rehmannia granule can improve the neurobehavioral performance of the PD rats with LID.
