补中益气汤“要药”的配伍对脾虚大鼠胃肠动力影响的机制研究 被引量:27

Mechanisms Study of Key Herb Compatibility in Buzhong Yiqi Decoction on Gastrointestinal Motility in Rats with Spleen Deficiency

摘要 目的:探讨具有不同升清作用的补中益气汤"要药"升麻柴胡的配伍对脾虚大鼠胃肠动力影响的药效学差异及作用机制。方法:大鼠随机分为正常组、模型组、莫沙必利组、补中益气汤组、全方去升柴组、全方去升麻组、全方去柴胡组、升麻柴胡组、升麻组、柴胡组,除正常组外,其余各组大鼠均采用三因素复合法复制脾虚胃肠动力障碍模型。给药14 d后,测定各组大鼠的胃排空率和小肠推进率,ELISA法检测各组大鼠血浆胃动素(motilin,MTL),血清胃泌素(gastrin,GAS)和血清肌酸激酶(CK)含量。结果:(1)模型组大鼠的胃排空率和小肠推进率均明显低于正常组(P〈0.01);莫沙比利组、补中益气汤组、全方去柴胡组、升麻柴胡组、升麻组的胃排空率和小肠推进率均明显高于模型组(P〈0.01),柴胡组的胃排空率和小肠推进率也显著高于模型组(P〈0.05),而全方去升柴组大鼠的胃排空率和小肠推进率与模型组比较差异均不显著(P〉0.05);全方去升麻组大鼠的胃排空率与模型组比较显著升高(P〈0.01),但其小肠推进率差异不显著(P〉0.05)。(2)与正常组相比,模型组血浆MTL和血清GAS含量明显下降(P〈0.01),血清CK含量显著升高(P〈0.01);莫沙必利组、补中益气汤组、全方去升柴组、全方去柴胡组、升麻柴胡组、升麻组、柴胡组血浆MTL相比模型组显著升高(P〈0.01,P〈0.01,P〈0.05,P〈0.01,P〈0.01,P〈0.05,P〈0.05),补中益气汤组、全方去升柴组、全方去升麻组、全方去柴胡组、升麻柴胡组、升麻组、柴胡组血清GAS明显上升(P〈0.01),补中益气汤组、全方去柴胡组、升麻柴胡组、升麻组、柴胡组大鼠血清CK明显下降(P〈0.01);全方去升麻组大鼠血清CK明显下降(P〈0.01),而其血浆MTL无明显变化(P〉0.05);全方去升柴组大鼠血清CK无明显变化(P〉0.05),莫沙必利组大鼠血清GAS和血清CK均无明显变化(P〉0.05,P〉0.05)。结论:升麻对全方的贡献度大于柴胡,证实了"柴胡为升麻之辅佐"的论述;补中益气汤"要药"配伍的促胃肠动力作用主要是通过调节胃肠激素水平实现的;补中益气汤"要药"的配伍对过度运动造成的骨骼肌损伤可能具有干预作用。 Objective:This study is aimed at uncovering the difference in pharmacodynamics and mechanisms of Key herbs Shengma and Chaihu compatibility in Buzhong Yiqi Decoction. Method: Rats were randomly divided into Control group( CTG), Model group (MODG) , Mosapride group ( MPG), Buzhong Yiqi Decoction group ( BZYQDG), Buzhong Yiqi Decoction without compatibility of Shengma - Chaihu group ( BZYQUSCG), Buzhong Yiqi Decoction without compatibility of Shengma group ( BZYQUSG), Buzhong Yiqi Decoction without compatibility of Chaihu group ( BZYQUCG), Shengma- Chaihu group( SCG), Shengma group( SG), Chaihu group(CG). All groups were duplicated by method re-coded in literature. After 14 days of administration, gastric emptying rate and small intestinal propulsion rate were detected. ELISA was applied to detect content of MTL in serum, GAS and CK in plasma. Result : ( 1 ) Compared to that of CTG, gastric emptying rate and small intestinal propulsion rate of rats in spleen deficiency model group decreased significantly (P 〈0. 01 ). Compared to that of MODG, gastric emptying and small intestinal propulsions of MPG, BZYQDG, BZYQUCG, SCG, SG increased significantly (P 〈 0. 01 ). The gastric emptying and small intestinal propulsion of CG increased significantly(P 〈 0. 05 ), while there was no obvious change in BZYQUSCG (P 〉 0. 05). Compared to that of MODG, gastric emptying rate in BZYQUSG increased significantly (P 〈 0. 01 ), while there was no obvious change in small intestinal propulsion rate (P 〉 0. 05 ). (2)Compared to that of CTG, MTL content in MODG plasma and GAS content in MODG serum decreased significantly (P 〈 0. 01 ) , while CK content in MODG serum increased significantly (P 〈 0. 01 ). Compared to that of MODG, MTL content in plasma and GAS content in serum increased significantly (P 〈 0. 01, P 〈 0. 01, P 〈 0. 01, P 〈 O. 05, P 〈 0. 05 ; P 〈 0. 01 ) , while CK content in serum decreased significantly ( P 〈 0. 01) in BZYQDG, BZYQUCG, SCG, SG, CG. MTL content in plasma and GAS content in serum in BZYQUSCG increased significantly (P 〈 0. 05, P 〈 0. 01 ) , and there was no significant change on CK in serum (P 〉 0. 05 ). GAS con- tent in serum in BZYQUSG increased significantly ( P 〈 0. O1 ) and CK content in serum decreased significantly ( P 〈 0. 01 ) , and there was no obvious change on MTL in plasma (P 〉 0. 05). MTL content in plasma in MPG increased significantly (P 〈 0. 01 ) , while there was no obvious change on GAS and CK in serum (P 〉 0. 05 ). Conclusion: Shengma contributes more than Chaihu to the effect of Buzhong Yiqi Decoction, which confirms the arguments of ZHANG Shanlei. The furtherance of gastrointestinal motility of key herb compatibility in Buzhong Yiqi Decoction might be achieved by regulating gastrointestinal hormone levels. It might be of some protective effect for Buzhong Yiqi Decoction on skeletal muscle injury due to over exercise.
