摘要
以新鲜的鲽鱼为原料,采用酸法从其黑皮和白皮中提取胶原蛋白,并对黑皮和白皮酸溶性胶原蛋白的理化特性和功能性质进行比较研究,为鲽鱼的开发利用提供一些理论依据。通过紫外光谱、傅里叶红外光谱扫描初步研究黑皮和白皮酸溶性胶原蛋白的结构特征,并对其色泽、凝胶强度、吸湿性和保湿性进行研究。紫外光谱扫描结果表明,黑皮和白皮ASC分别在230 nm和230.5 nm处有强吸收峰,均符合胶原蛋白的特征。傅里叶红外光谱扫描结果表明,黑皮ASC在3 338,2 929,1 647,1 535,1 242 cm^(-1)处均有吸收峰,而白皮ASC在3 334,2 931,1 654,1 541,1 238 cm^(-1)处也有吸收峰。由此证实黑皮和白皮ASC都存在酰胺A、酰胺B、酰胺Ⅰ、酰胺Ⅱ、酰胺Ⅲ,均保持胶原蛋白三维螺旋结构的完整性。通过色泽、凝胶强度的测定,结果表明,黑皮和白皮ASC的白度分别为66.80±1.29,73.31±1.95,凝胶强度分别为(523.09±4.85)g·mm和(431.34±2.15)g·mm。保湿性和吸湿性研究表明,黑皮ASC的保湿率明显高于白皮ASC保湿率(P<0.05),而黑皮和白皮ASC吸湿率差异不显著(P>0.05)。以上结果表明,从鲽鱼不同部位提取的ASC结构和功能特性有一定的差别。
Fresh flounder was used as the raw materials in this study, the black and white skin acid-soluble colla- gens were respectively extracted from black and white skin of flounder, of which physiochemical characteristics and func- tional properties were compared and analyzed. Structure characteristics of acid-soluble collagens of black and white skin were preliminarily studied by ultraviolet spectrum and Fourier infrared spectrum scanning. Meanwhile, Color, gel strength, hygroscopicity and moisture retention were also studied. Ultraviolet spectrum scanning revealed that there was a strong absorption peak at 230 nm in black skin acid-soluble collagen and a strong absorption peak at 230.5 nm in white skin acid-soluble collagen, which demonstrated the two kinds of collagens were in accordance with the characteristics of the collagen. Black and white skin acid-soluble collagens had absorption peaks at 3 338, 2 929, 1 647, 1 535, 1 242 cm-1 and 3 334, 2 931, 1 654, 1 541, 1 238 cm-1 respectively by analyzing IR spectra, which proved the existences of amide A, amide B, amide Ⅰ , amide Ⅱ, amide Ⅲ, confirming that triple helical conformation of the two kinds of colla- gens were reserved completely. The whiteness value and gel strength of black and white skin acid-soluble collagens re- spectively were 66.80 ±1.29, 73.31 ±1.95, 523.09 ±4.85 g.mm and 431.34 ±2.15 g.mm. Moisture retention and hygro- scopicity indicated that moisture-retention ability of black skin acid-soluble collagen was significantly higher than white skin acid-soluble collagen (P〈0.05). While, there was not significant for black and white skin acid-soluble collagens in moisture absorption rate (P〉0.05). These results described above showed that different extraction materials had certain dif- ference on the structure and functional characteristics of acid-soluble collagens from flounder skin.
