摘要
目的探讨间充质干细胞(MSC)过表达神经生长因子(NGF)对神经元缺糖缺氧(OGD)模型中神经元损伤的修复作用机制。方法分离和培养大鼠MSC并过表达NGF,收取MSC蛋白,Western blot法检测NGF的过表达效果;同时分离培养大鼠原代神经元,制备大鼠神经元OGD模型,然后将MSC培养上清液加入大鼠神经元OGD模型中,TUNEL染色法检测神经元凋亡情况,Western blot法检测保护性信号通路磷酸化Akt(p-Akt)表达情况。结果 Ad NGF腺病毒感染的MSC培养上清液可明显上调原代神经元p-Akt表达水平,过表达NGF后的MSC培养上清液可以进一步修复原代神经元损伤,抑制原代神经元凋亡。结论过表达NGF的MSC可修复神经元损伤,NGF联合MSC移植治疗神经元损伤或为临床一种新思路。
Objective To investigate the neuroprotective mechanism of overexpression of nerve growth factor(NGF)in mesenchymal stem cells(MSC)by using oxygen-glucose deprivation(OGD)model.MethodsRat MSCs were isolated and cultured,the overexpressed NGF was detected by Western blot.The rat primitive neurons were cultivated and neurons OGD model was prepared.MSC culture supernatant was added to the OGD model;the neuron apoptosis was detected with TUNEL staining,the expression of Akt protein and phosphorylation was detected with Western blot.ResultsThe supernatant of MSC culture infected by Ad NGF significantly increased the level of phosphorylated Akt.The culture supernatant of MSC overexpressing NGF protected neurons from apoptosis,promoted the recovery of central nervous function and alleviated central nervous system damage.Conclusion NGF combined with MSC transplantation provides a new way to treat central nerve injury and complications.
