十六味冬青散对COPD大鼠血清炎性因子及肺脏凋亡蛋白的影响

摘要 目的分析十六味冬青散对COPD大鼠血清炎性因子及肺脏凋亡蛋白的影响机制。方法选取60只清洁型的SD雌性大鼠,按照随机原则分成5组:A组COPD模型组、B组十六味冬青散高剂量组、C组十六味冬青散低剂量组、D组氨溴索治疗组、E组健康大鼠组。应用免疫组化法检测并比较各组肺脏中的相关凋亡蛋白(Bcl-2、Bax、Caspase-3)的浓度。采用血清酶联免疫吸附法(ELISA)检测并比较各组大鼠血清炎性因子(IL-6、IL-8、TNF-α)的表达。结果 A组炎性反应水平最高,E组炎性反应水平最低。C组炎性反应水平低于A组,但差异无统计学意义(P>0.05)。B组和D组炎性反应水平低于A组和C组,且D组研究反应水平低于B组,差异均有统计学意义(P<0.05)。A组Bax、Caspase-3水平最高,Bcl-2、Bcl-2/Bax水平最低,E组Bax、Caspase-3水平最低,Bcl-2、Bcl-2/Bax水平最高。B组和D组的Bax、Caspase-3水平低于A组和C组,Bcl-2、Bcl-2/Bax水平高于A组和C组,差异均有统计学意义(P>0.05)。A组和C组的Bax、Caspase-3、Bcl-2、Bcl-2/Bax水平比较差异无统计学意义(P>0.05)。B组和D组Bax、Caspase-3、Bcl-2、Bcl-2/Bax水平比较差异无统计学意义(P>0.05)。结论 COPD的起病原因可能与肺部炎症及肺脏细胞的凋亡有关,十六味冬青散在COPD肺组织治疗中可以起到抗炎、抗细胞凋亡的作用,其具体机制可能是调节凋亡途径中的Bax、Bcl-2以及Caspase-3等相关蛋白的表达,阻止细胞炎性因子的表达,从而起到调节肺功能的作用。 Objective To analysis the effect of 16 flavor holly powder on the serum inflammatory factor and and lung apoptotic protein in patients with COPD rats. Methods Selected 60 cases clean SD female rats. According to the random principle,60 cases rats were divided into 5 groups: group A COPD model group,group B sixteen flavors of winter green powder high dose group,group C sixteen flavors of winter green powder low dose group,group D ambroxol treatment group,group E healthy rats group. Immunohistochemistry was used to detect and compare the concentrations of related apoptotic proteins( Bcl-2,Bax,Caspase-3) in the lungs of 5 groups. The serum inflammatory factors( IL-6,IL-8,TNF-α) were detected by serum enzymelinked immunosorbent assay( ELISA). Results Group A had the highest level of inflammatory response and group E had the lowest level of inflammatory response. The level of inflammatory response in group C was lower than that in group A,but the difference was no statistically significant( P 〉0. 05). The inflammatory response level of group B and group D was lower than that of group A and group C,and the response level of group D was lower than that of group B,the difference was statistically significant( P 〈0. 05). The levels of Bax and Caspase-3 were the highest in group A,the lowest in Bcl-2 and Bcl-2/Bax,the lowest in Bax and Caspase-3 in group E,and the highest in Bcl-2 and Bcl-2/Bax. The levels of Bax and Caspase-3 in group B and group D were lower than those in group A and C. The levels of Bcl-2 and Bcl-2/Bax were higher than those in group A and C. The difference was statistically significant( P 〉0. 05). There was no significant difference in the levels of Bax,Caspase-3,Bcl-2 and Bcl-2/Bax between group A and group C( P 〉0. 05). There was no significant difference in the levels of Bax,Caspase-3,Bcl-2 and Bcl-2/Bax between group B and group D( P〉 0. 05). Conclusion The onset reason may be related to lung inflammation and apoptosis of lung cells,Sixteen flavors of holly powder can play an anti-inflammatory and anti-apoptotic role in the treatment of COPD lung tissue. The specific mechanism may be to regulate the expression of Bax,Bcl-2 and related proteins such as Caspase-3 in the apoptotic pathway. Prevents the expression of cellular inflammatory factors,thereby regulating lung function.
