摘要
目的建立一测多评(QAMS)法同时测定广金钱草中5种黄酮成分(夏佛塔苷、维采宁-2、异夏佛塔苷、异荭草苷、异牡荆苷),研究不同生长期不同荫蔽度对广金钱草生长和药效成分含量的影响。方法设置0(不遮阴)、50%、75%、90%4个不同荫蔽度,对比不同生长期各处理组广金钱草的产量及形态特征,并通过QAMS测定各处理组的药效成分。结果维采宁2、异夏佛塔苷、异荭草苷、异牡荆苷与内参物夏佛塔苷的相对校正因子分别为1.222,1.275,1.402,1.554,QAMS法与外标法余弦相似度均大于0.995,相关性良好。遮阴处理下的广金钱草产量显著减少,生长形态和活性成分含量有差异。结论 QAMS法可用于广金钱草中5种黄酮成分的含量测定。广金钱草采收期可考虑在花期,光照有利于花期广金钱草黄酮含量的积累,在花期过后,适当的遮阴可减缓黄酮成分的代谢。
Objective To establish a Quantitative Analysis of Multi-components with Single Marker(QAMS) method for simultaneous determination of five flavonoids in Desmodium styracifolium(HDS)and discuss the influence of light intensities on growth of the herb and the contents of active constituents of HDS.Methods The contents of five flavonoids from samples grown in different shading sheds(0,50%,75%,90%)were determined by external standard method(ESM)and QAMS.Results There is no significant difference between the ESM and QAMS in the quantitative results of five constituents of HDS.Light shading caused significant effect on the growth and the contents of active constituents in HDS and decreased production.The RCFs of vicenin-2,isoschaftoside,isoorientin,isovitexin were 1.222,1.275,1.402,1.554.Conclusion The QAMS method was verified to be an alternative and promising method for multi-component determination of HDS.Flowering time was the most appropriate harvest time of HDS.Sufficient illumination is necessary for HDS at flowering period.After the flowering period,proper shading can slow down reducing of the contents of flavonoids.
