摘要
上皮间质转化(Epithelial mesenchymal transition,EMT)是肿瘤发展的重要过程,肿瘤细胞与良性细胞间的信息传递能够促进肿瘤的发展。由肿瘤细胞的培养液获取的条件培养基(Conditioned medium,CM)中富含细胞分泌的众多因子,这些细胞因子能够改变正常细胞的行为。将基质浸润性膀胱癌细胞YTS-1获取的条件培养基(YCM)用于培养膀胱上皮细胞HCV29后,发现处理后的HCV29细胞高表达间质细胞标志蛋白fibronectin和N-cadherin,同时细胞的迁移和增殖能力明显增强,凋亡能力减弱。同时,通过基质辅助激光解吸飞行时间质谱以及凝集素芯片技术对Y-CM处理前后的HCV29细胞进行了完整的糖链表达谱解析,Y-CM处理后细胞高甘露糖型N-糖链表达量明显减少,而杂合型、复合型以及岩藻糖和唾液酸型N-糖链表达量均有不同程度的提升。综上,研究揭示了恶性膀胱癌细胞获取的条件培养基能够诱导膀胱上皮细胞发生恶性转变,发生EMT转化并改变细胞的糖链表达谱。
Epithelial mesenchymal transition (EMT) is a key process in cancer development. Communication between cancer cells and normal cells may facilitate cancer progression. Conditioned medium (CM) from cultured cancer cells containing secreted factors may affect behaviors of normal cells.In this study, normal bladder epithelial HCV29 cells were cultured with CM from malignant bladder cancer YTS-1 cells (Y-CM). In comparision with control cells, HCV29 treated with Y-CM showed the increased expression of mesenchymal markers fibronectin and N-cadherin enhanced cell proliferation, migration,and inhibition of cell apoptosis. Furthermore, N-glycan profiles of Y-CM treated and control HCV29 were compared by MALDI-TOF-MS and lectin microarray. The treated cells showed lower levels of high-mannose type N-glycan structures,and higher levels of complex-type, hybrid-type, fucose-type and sialic acid-type structures. Taken together, our findings indicated that CM derived from bladder cancer cells induced an EMT-like process in normal bladder epithelial cells and altered their N-glycan profile.
