摘要
目的:观察甘草附子汤对类风湿关节炎大鼠血清和滑膜组织中ASIC3和HIF-1α表达影响,探讨甘草附子汤治疗RA的相关机制。方法:40只SPF级Wistar大鼠随机分为4组,每组10只,除正常组外,其余组等量的不完全氟氏佐剂和牛Ⅱ型胶原乳化剂造模。甘草附子汤(9.75g/kg),醋酸地塞米松(7.5 mg/kg),连续灌胃20 d。分别拍摄大鼠足跖X射线,用ELISA检测大鼠血清中HIF-1α和ASIC3表达,免疫组化法检测关节滑膜组织中HIF-1α和ASIC3蛋白表达。结果:模型组大鼠血清和滑膜组织中ASIC-3和HIF-1α含量明显高于正常组(P<0.01),甘草附子汤组大鼠滑膜组织和血清中ASIC-3、HIF-1α含量明显低于模型组(P<0.01)。结论:甘草附子汤可以降低RA模型大鼠血清和滑膜组织中ASIC3、HIF-1α含量,可能是该方治疗RA的作用机制之一。
Objective: To observe the effect of Licorice and Aconite decoction(甘草附子汤) on the expression of ASIC3 and HIF-1α in serum and synovial tissue of rats with rheumatoid arthritis(RA), and to explore and elaborate the related mechanism of Licorice and Aconite decoction in the treatment of RA. Methods: The40 SPF Wistar rats were randomly divided into 4 groups with 10 rats in each group. Except normal group, the other groups were modeled with incomplete Freund's adjuvant and bovine typeⅡcollagen emulsifier. Licorice and Aconite decoction 9.75 g/kg, dexamethasone acetate 7.5 mg/kg, continuous gavage for 20 days. X-rays of the rat's foot were taken respectively. The expression of HIF-1α and ASIC3 in the serum of the rats was detected by Elisa. The expression of HIF-1α and ASIC3 in the synovial tissue was measured by immunohistochemistry.Results: Compared with the normal group, the expression of ASIC-3 and HIF-1α in the serum and synovial tissue of the model group were significantly higher(P〈0.01). Compared with the model group, the expression of ASIC-3 and HIF-1α in the synovial tissue and serum of the Licorice and Aconite decoction was significantly lower(P〈0.01). Conclusion: Licorice and Aconite decoction can reduce the content of ASIC3 and HIF-1α in rat serum and synovial tissue of RA model rats, which may be one of the mechanisms of the treatment of RA.
