摘要
目的研究Zeste基因增强子同源物2(EZH2)对肺癌细胞中沉默抑制子1(ROS1)表达及细胞上皮-间质转化的影响。方法 Western blot检测肺癌细胞中EZH2表达;过表达EZH2后,RT-PCR和Western blot检测肺癌细胞HCC461中EZH2表达水平;Transwell实验检测细胞侵袭迁移能力变化;Western blot检测过表达EZH2对ROS1、E-cadherin、Snail、vimentin表达影响;Western blot检测53例肺癌组织和42例癌旁组织中EZH2与ROS1表达水平并分析其相关性。结果肺癌细胞中EZH2均高表达;肺癌细胞HCC461过表达EZH2后ROS1表达上调,上皮标记物E-cadherin下调,间质标记物Snail、vimentin表达上调,细胞侵袭转移能力增加;肺癌组织中EZH2、ROS1均显著高表达,且两者表达呈显著正相关。结论 EZH2可介导肺癌细胞中ROS1表达上调来促进肺癌细胞上皮-间质转化。
Objective To investigate the effect of Zeste gene enhancer homologue 2 (,EZH2) on repressor of silencing 1 (ROS1) expression and cell epithelial-mesenchymal transition (EMT)in lung cancer cells. Methods Western blot was used to detect the expression of EZH2 in lung cancer cells. After overexpression of EZH2, the expression of EZH2 in lung cancer cell line HCC461 was detected by RT-PCR and western blot. Transwell assay was used to detect the invasion and migration. The ROS1, E-cadherin, Snail and vimentin expression were detected by western blot. Western blot was used to explore the relations between EZH2 and ROS1 protein levels. Results EZH2 is highly expressed in lung cancer cells. Overexpressing of EZH2 in HCC461 cells lead to the upregulation of ROS1 and mesenchymal markers Snail and vimentin, while theepithelial marker E-cadherin was down-regulated ; the ability of cell invasion and migration was increased. Overexpression of EZH2 was correlated with the upregulation of ROS1 in human lung cancer tissues. Conclusion EZH2 could upregulate the expression of ROS1 in lung cancer cells to promote the epithelial-mesenchvmal transition of lung cancer cells.
