补肾化痰法经APN信号通路对肥胖PCOS大鼠GC增殖的调控研究 被引量:6

Influence of Granulosa Cells Multiplication of Method of Kidney Nourishing and Phlegm Removing Through APN Pathway in Obese PCOS Rats

摘要 目的:观察补肾化痰法经APN通路对肥胖PCOS大鼠卵巢GC增殖的调节作用研究。方法:采用来曲唑加高脂建立肥胖PCOS大鼠模型,补肾化痰复方治疗后,取卵巢组织行HE染色,免疫组化法检测AdipoR1/AdipoR2表达,realtime-PCR检测GC中p38MAPK、smad1/3、CCND2的mRNA表达水平。结果:与模型对照组比较,中药高、中剂量组卵巢GC中AdipoR1/2的表达增高(P〈0.05);中药高、中剂量组卵巢颗粒细胞p38MAPK、smad1/3、CCND2表达量均增高(P〈0.05)。结论:补肾化痰法可能通过APN信号通路促进GC增殖,进而改善肥胖PCOS卵泡发育障碍,达到治疗作用。 Objective: To observe of the influence of granulosa cells(GC) multiplication of the method of kidney nourishing and phlegm removing(KNPR) through the APN pathway in obese PCOS rats. Methods: The rats were treated with the method of KNPR after modeling by Letrozole and high-fat. The ovarian tissues were taken out for HE staining,and the expressions of AdipoR1 and AdipoR2 were detected by immunohistochemical staining method,and the mRNA expressions of p38 MAPK,smad1/3 and CCND2 were detected by realtime-PCR. Results: Compared with model group,the AdipoR1/2 of high dose group(HDG) and middle dose group(MDG) was increased(P〈0. 05). The mRNA expressions of p38 MAPK,smad1 and smad3 of HDG and MDG were increased(P〈0. 05). Conclusion: The method of KNPR may promote the multiplication of GC by the APN pathway and improve the follicle developmental disorder of the obese PCOS to achieve the therapeutic effect.
