果胶对小鼠抗氧化能力的影响及DNA损伤的干预 被引量:2

The role of Pectin influencing antioxidant capacity and intervening DNA damage in mice

摘要 目的探究杨梅精品"黑高峰"中果胶对高胆固醇模型小鼠血浆胆固醇及其相关血脂生化指标及对小鼠抗氧化能力和DNA损伤的影响。方法将48只健康初断乳C57BL小鼠,随机分为对照组、低浓度果胶组、高浓度果胶组。各组饲料果胶含量分别为0、70、200(mg/kg)。制作由0、6、12、18μmol/L不同浓度H_2O_2诱导的DNA损伤兼高胆固醇模型。实验期限为60d。实验结束后采血,采用试剂盒检测血清谷胱甘肽过氧化物酶(GSH)和丙二醛(MDA)水平,并应用单细胞凝胶电泳法测定淋巴细胞DNA的损伤情况。结果与对照组比较,低浓度果胶组、高浓度果胶组血清GSH和MDA含量差异有统计学意义,P <0. 01。低浓度果胶组和高浓度果胶组小鼠血清总胆固醇TC、载脂蛋白B(apo B)、甘油三酯(TG)的含量都有明显的降低,尤其以高浓度果胶组最为显著,同时,两组小鼠的胆汁酸排泄量增加。但对载脂蛋白AⅠ(apoA-Ⅰ)、高密度脂蛋白-胆固醇(HDL-C)水平影响不明显。结论果胶可降低血浆胆固醇水平。对于健康幼年小鼠血清GSH及脂质过氧化产物MDA含量有显著影响,对H_2O_2诱导的DNA损伤表现出明显的保护作用。 Objective To explore the effects of pectin in the " black peak" of Yangmei boutique on plasma cholesterol and its related blood lipid biochemical indexes in high cholesterol model mice and its effects on antioxidant capacity and DNA damage in mice. Methods 48 healthy male weanling C57BL mice were randomly divided into control group, low concentration group and high concentration group of pectin. The content of pectin in each group was 0.70.200 (mg / Kg), respectively. Different concentrations of H2O2(0. 6, 12, 18 H2O2 ) were used to induce DNA damage and high cholesterol model. The experimental period was 60d. Bloods amples were collected at the end of the experiment, the levels of serum glutathione peroxidase (GSH) and malondialdehyde (MDA) were detected by kit, and the damage of lymphocyte DNA was detected by single cell gel electrophoresis. Results Compared with the control group, the serum levels of GSH and MDA in low concentration pectin group and high concentration pectin group were significantly different ( P = 0. 4361 ), with statistical significance. The low concentration group and high concentration of total cholesterol, bile acid excretion in two groups of mice increased. But the effect of apolipoprotein A I ( apoA - I) and high density lipoprotein cholesterol (HDL - C) level was not obvious. Conclusion Pectin could reduce plasma cholesterol level, it had a significant effect on serum GSH and lipid peroxidation MDA content in healthy young mice, and had obvious protective effect on H2O2 -induced DNA damage.
