摘要
对鸡群中感染的Ⅰ群禽腺病毒hexon基因进行分型与序列分析,以期明确Ⅰ群禽腺病毒基因型的分布和hexon基因变异情况。参考GenBank上FAdV基因序列,设计2对引物,采用套式PCR技术扩增临床病料中Ⅰ群禽腺病毒hexon基因,测序后获得了8株FAdV hexon基因片段。结果表明,构建系统进化树发现,其中HM、SXFS、LQQD、HXGB、GSPL、DYQY和FP共7株属于FAdV-4型,而GS株属于FAdV-1型。7株FAdV-4型毒株核苷酸同源性在99.9%~100%之间,推导的氨基酸序列同源性均为100%,提示这7株FAdV-4型流行毒株高度同源。FAdV-1型的GS与CELO株核苷酸、推导的氨基酸序列同源性均为99.2%,提示GS与CELO株亲缘关系较近。在鸡群中发现存在FAdV-1型和FAdV-4型Ⅰ群禽腺病毒,各血清型毒株之间高度同源,毒株遗传稳定性高。
The genotyping and sequence analysis of the hexon gene of group I fowl adenovirus(FAdV) infected in chickens were performed in order to clarify the distribution of genotypes of group I FAdV and the mutation of hexon gene.Two pairs of primers were designed according to the FAdV gene sequence in GenBank.The hexon gene of group I FAdV was amplified by nested PCR,eight fragments of FAdV hexon gene were obtained after sequencing. The results showed that HM, SXFS, LQQD, HXGB, GSPL, DYQY and FP strains belong to FAdV-4,and GS strain belongs to FAdV-1;The nucleotide homology of the isolates of FAdV-4 was between 99.9% and 100%, and the deduced amino acid sequence homology was 100% ,which indicated that the seven strains of FAdV-4 were highly homologous.The homology of GS and CELO strains of FAdV-1 was 99.2% ,which indicated that the relationship between GS and CELO was higher.FAdV-1 and FAdV-4 were found in flocks.The strains were highly homologous among serotypes, and the strains had high genetic stability.
