摘要
目的探讨高山灌木苏嘎纯提取液是否具有抗氧化应激作用。方法采用MTT法检测终质量浓度分别为100、50、25 mg/L的苏嘎纯提取液对角朊细胞存活率影响;利用体外培养人角朊细胞系,给予H2O 2处理造成细胞氧化应激模型,采用100mg/L的苏嘎纯提取液预先作用角朊细胞,MTT法检测细胞存活率,并用Annexin V的方法检测细胞凋亡指数。结果通过MTT法检测,发现浓度为100mg/L以下的苏嘎纯提取液对细胞生长没有明显损伤;采用100mg/L的苏嘎纯提取液预处理角朊细胞,使该细胞存活明显增加;经过Annexin V细胞凋亡检测,发现100mg/L的苏嘎纯提取液能够抑制H_2O_2诱导的角朊细胞凋亡。结论苏嘎提取取液具有抑制角朊细胞凋亡,拮抗氧化应激的作用。
Objective: To investigation the induction of keratinocytes apoptosis by inhibiting oxidative stress in crude suga extract from Alpine shrub. Methods: MTT assay was used to detect the effect of crude suga extract with different concentration(100 mg/L, 50 mg/L, 25 mg/L) on the survial rate of keratinocytes. The artificial keratinocyte was cultivated and oxidized for establishing oxidative stress model. The 100 mg/L of crude suga extract was dripped onto oxidative stress model. Then the survial rate of keratinocytes was determined by MTT assay, and the apoptosis was determined by Annexin V. Results: In this study, all crude suga extract had no obvious effect on keratinocytes growth. As oxidative stress model, increases in survival of keratinocytes was determined, and keratinocytes apoptosis was inducted by inhibititing oxidative stress. Conclusions: The crude suga extract can inhibit keratinocytes apoptosis.
