摘要
目的建立局灶节段性肾小球硬化(FSGS)型阿霉素肾病大鼠模型。方法 72只SD大鼠随机分为假手术组(A组,12只)和FSGS模型组(B组,60只)。B组于左肾切除术后1周静脉注射阿霉素3mg/kg,4周后再均分为四组,分别静脉注射不同剂量阿霉素0、1、2、3mg/kg。采用脲酶UV法检测BUN和SCr水平,邻苯三酚红/钼酸盐比色法检测24-h尿蛋白定量,并于光镜和电镜下观察肾组织病理形态学改变。结果与A组相比,B组阿霉素注射4周后的24-h尿蛋白增加(P<0.01),BUN和SCr水平略微升高(P>0.05),肾组织病理出现肾小球系膜细胞及基质轻度增生,足突细胞融合。不同剂量阿霉素注射8周后的24-h尿蛋白和BUN水平均较A组升高(P<0.01),肾组织病理出现肾小球毛细血管腔狭窄、闭塞,表现不同程度的FSGS,足突细胞融合和毛细血管内皮细胞增生;其中,阿霉素2mg/kg剂量组30%~50%肾小球有不同程度的硬化,呈典型的局灶、节段性分布。结论阿霉素首次静脉注射3mg/kg,间隔4周再注射阿霉素2mg/kg,可建立FSGS型肾病大鼠模型。
Objective To establish a nephropathy model with focal segmental glomurular sclerosis(FSGS)in rats induced by adriamycin.Methods Seventy-two SD rats were randomly divided into two groups of A(sham operated,12 rats)and B(FSGS model,60 rats).The rats in group B were intravenously injected with adriamycin 3 mg/kg in one week after left nephrectomy,and then equally divided into four groups by injecting different doses of adriamycin 0,1,2 and 3 mg/kg after four weeks,respectively.The levels of BUN and SCr were detected by urease UV assay and 24-hour urine protein was measured by pyrogallol red/molybdate method.The pathomorphology of renal tissues was observed under light microscope and electron microscope.Results Compared with group A,24-hour urine protein was increased(P〈0.01),BUN and SCr were slightly increased(P〉0.05),and renal pathology showed mildly hyperplastic glomerular mesangial cells and matrix and fused podocytes in group B after four weeks.The level of BUN and 24-hour urine protein after injected with different doses of adriamycin were all higher than those in group A(P〈0.01),and renal pathology showed narrow glomerular capillary,different degrees of FSGS,fused podocytes and hyperplastic capillary endothelial cells after eight weeks.Especially,30%-50% glomerulus appeared different degrees of sclerosis with typical focal and segmental distribution after injection of adriamycin 2 mg/kg.Conclusion FSGS nephropathy model in rats can be successfully induced by first injection of adriamycin 3 mg/kg,which is followed by injection of adriamycin 2 mg/kg at an interval of four weeks.
