摘要
为了研究肽硫转移酶mu1(GSTM1)基因与超氧化歧化酶3(SOD3)的基因多态性,探索影响草原红牛剩余采食量性状的候选基因,试验测定了96头草原红牛血液基因组中GSTM1基因的编码区和SOD3基因的5'UTR区,并进行DNA混池、多态性、等位基因数、核苷酸多态位点、氨基酸多态位点分析,以探究GSTM1基因与SOD3基因在草原红牛基因组中的多态性表现。结果表明:草原红牛GSTM1基因编码区中704位存在C-T突变,这种突变是错义突变,氨基酸发生变化由丙氨酸变成缬氨酸,可能会使蛋白质结构发生变化。在SOD3基因的5'UTR区存在两个SNP位点,分别在71位发生G-A突变和117位C-T突变,5'UTR区上基因的突变可能导致基因表达发生变化。在遗传水平上草原红牛剩余采食量性状的差异可能是由多个基因多态性造成的。
The aim of the present study was to investigate the polymorphism of GSTM1 gene and SOD3 gene,and explore the candidate genes affecting the residual feed intake( RFI) traits of Red Steppe cattle. The coding region of GSTM1 gene and the 5'UTR region of SOD3 gene in blood DNA of 96 Red Steppe cattle were measured,and the DNA mixed pool,polymorphism analysis,allele number,nucleotide polymorphic loci and amino acid polymorphic loci were analyzed in order to explore the polymorphism of GSTM1 gene and SOD3 gene in genome of Red Steppe cattle. The results showed that there were 704 C-T mutations in the coding region of GSTM1 gene in Red Steppe cattle. This mutation was a missense mutation. The amino acids changed from alanine to valine,which might change the structure of protein. There were two SNP sites in 5' UTR region of SOD3 gene,which located 71( G-A mutations) and 117( C-T mutations). Mutations in the 5'UTR region might lead to changes in gene expression. At the genetic level,the difference in the RFI traits of Red Steppe cattle might be due to multiple gene polymorphisms.
