摘要
目的:初步研究白及茎秆醇提物的化学成分和抗肿瘤作用,更好地开发利用白及药用资源。方法:采用乙醇回流法提取白及茎秆和块茎醇提物,对醇提物进行化学成分分析。不同剂量白及茎秆醇提物处理体外培养的肺腺癌细胞(A549),用MTS法检测A549细胞的增殖情况,划痕法检测A549细胞的迁移率,Hoechst-PI染色法检测白及茎秆醇提物对A549细胞凋亡的影响,RT-PCR法检测白及茎秆醇提物对A549细胞Bcl-2、Bax和ICAM-1基因mRNA表达的影响。结果:白及茎秆醇提物化学成分同块茎类似,并能抑制A549细胞的增殖,且存在剂量依赖关系。随着药物浓度的增加,Bcl-2、ICAM-1 mRNA表达下调,Bax mRNA表达上调。结论:白及茎秆醇提物可通过抑制A549细胞的生长增殖,影响细胞凋亡相关基因的表达发挥抗肿瘤作用。白及茎杆有潜力成为块茎的补充资源。
Objective: To study the chemical compositions and the anti-tumor effect of alcohol extract from Bletilla striata stem parts,in order to develop and utilize Bletilla striata medicinal resources better. Methods: By using the ethanol refluxing methods to extract the Bletilla striata's stem and tuber parts,we analyzed the chemical compositions of alcohol extracts. Using alcohol extracts of Bletilla striata stem parts,we treated A549 cells in vitro in different doses. The proliferation of A549 cells would be detected by MTS. The mobility of A549 cells would be detected by scratch method.Effects of alcohol extract from Bletilla striata stem parts on apoptosis of A549 cells by Hoechst-PI Staining. We detected Bcl-2,Bax and ICAM-1 mRNA expressions in alcohol extract from Bletilla striata stem parts by RT-PCR. Results:The chemical compositions of the alcohol extracts of Bletilla striata stem parts were similar to that from tubers which can inhibit the proliferation of A549 cells in a dose-dependent manner. With the increase of drug concentration,the expressions of Bcl-2 and ICAM-1 mRNA were down-regulated and the expression of Bax mRNA was up-regulated. Conclusion: The alcohol extracts for Bletilla striata stem parts can inhibit the growth and proliferation of A549 cells,affecting the expression of apoptosis-related genes which play an anti-tumor effect. Bletilla striata stem parts have the potential to become tubers supplementary resources.
