枸杞LbXEGIP1基因的克隆与生物信息学分析 被引量:1

摘要 内切葡聚糖酶抑制蛋白(XEGIP)在植物防御病原菌侵袭中扮演了重要角色。本研究以宁夏枸杞(Lycium barbarum L.)‘宁杞1号’为试验材料,采用RACE技术从枸杞花药cDNA中成功克隆了枸杞的XEGIP1基因,命名为LbXEGIP1,GenBank登录号为MG744343。结合生物信息学方法对LbXEGIP1基因编码的蛋白质进行理化性质、信号肽、亚细胞定位、蛋白结构以及系统进化等方面的预测分析。结果表明,该基因CDS序列全长1 290 bp,编码429个氨基酸,预测蛋白分子量为47.39 kD,等电点为8.59,是一个含有一个跨膜区和信号肽区域,脂溶性的稳定蛋白,它的亚细胞定位于细胞分泌途径中。LbXEGIP1属于天冬氨酸蛋白酶A1亚家族,在其N端和C端分别具有葡聚糖酶抑制子结构域。在其二级结构中无规则卷曲所占比例最高(35.66%),其次是延伸链(30.77%)。系统进化分析表明LbXEGIP1蛋白与甜椒中的XEGIP亲缘关系最近。枸杞XEGIP1基因的克隆及生物信息学分析,为后续研究该基因的功能以及枸杞病虫害的防治方面提供了依据。 Xyloglucan-specific endoglucanase inhibitor protein (XEGIP) plays a crucial role in plant defense against pathogenic bacteria. In this study, Ningxia wolfberry (Lycium barbarum L.) variety ‘Ningqi No. 1' was used as materials, and XEGIP1 gene was cloned from the cDNA ofwolfberry anther by RACE method, which was named as LbXEGIP1, with the accession number of GenBank of MG744343. Bioinformatics analysis was used to predict the physico-chemical properties, signal peptide, subcellular localization, protein structure and phylogenetic evolution of LbXEGIP1 encoded protein. The results showed that the full-length CDS was 1 290 bp, encoding 429 amino acids. The molecular weight of putative protein was 47.39 kD and its isoelectric point was 8.59, which was a lipid soluble stable protein containing a transmembrane and signal peptide regions, and its subcellular localization was in the cell's secretory pathway. LbXEGIP1 belonged to aspartic peptidase A1 subfamily, and its N and C terminals contained highly conserved xyloglucan inhibitor domain. In its secondary structure, the random coil accounted for the highest proportion (35.66%), followed by extended strands (30.77%). Phylogenetic analysis showed that LbXEGIP1 had the closest relationship with XEGIP in Capsicum annuum. Cloning and bioinformatics analysis ofXEGIP1 gene in wolfberry would provide reference for further research on the function of the gene and the disease resistance in wolfberry.
