摘要
目的研究川芎地上部分中3种活性成分的含量及其在生长过程中的变化。方法采用UPLC法分别测定川芎茎及叶中绿原酸、3,5-O-二咖啡酰奎宁酸、Z-藁本内酯的含量。采用Waters BEH C_(18)色谱柱(50 mm×2. 1 mm,1. 7μm),流动相为乙腈-0. 1%磷酸水溶液,梯度洗脱,流速0. 4 mL·min(-1),柱温35℃,检测波长325 nm。结果绿原酸、3,5-O-二咖啡酰奎宁酸、Z-藁本内酯的线性范围分别为0. 78~113. 64 ng(r=0. 9999)、0. 37~94. 38 ng(r=0. 9999)、0. 52~67. 08 ng(r=0. 9998),3个化合物的平均回收率分别为103. 54%、100. 44%、103. 15%,RSD分别为1. 35%、1. 98%、2. 91%。结论所用方法准确、简便、重复性好,适用于川芎地上部分中绿原酸、3,5-O-二咖啡酰奎宁酸、Z-藁本内酯的含量测定;川芎茎及叶中3种成分在生长过程中的变化趋势基本相同,约在五月中旬达到最高含量。
OBJECTIVE To determine the contents of three kinds of active component from the stems and leaves of Ligusticum chuanxiong during growth period. METHODS Contents of the three compounds were simultaneously determined by UPLC. A Waters BEH C_(18) column(50 mm × 2. 1 mm,1. 7 μm) was used with the mobile phase contain 0. 1% phosphoricacid and acetonitrile using a gradient program. The flow rate was 0. 4 mL·min(-1),with the column temperature of 35 ℃ and the wavelength of 325 nm. RESULTS The linear ranges of chlorogenic acid,3,5-O-dicaffeoylquinic acid and Z-ligustilide were 0. 78-113. 64 ng( r = 0. 9999),0. 37-94. 38 ng( r = 0. 9999) and 0. 52-67. 08 ng( r = 0. 9998),respectively. The average recoveries of three compounds were 103. 54%,100. 44% and 103. 15%,with the RSDs of 1. 35%,1. 98% and 2. 91%,respectively. CONCLUSION The method is simple,accurate and reproducible,and it is suitable for the content determination of the three kinds of active components from stems and leaves of L. chuanxiong. The maximum contents of the three compounds from both stems and leaves appeared in the middle of May with similar variation trend.
