摘要
利用CRISPR/Cas9基因组编辑技术对一个水稻穗发育相关基因OsD1进行了定点编辑。通过定点编辑载体的构建、水稻愈伤组织的转化、转基因植株的鉴定及突变类型的分析,获得了4株OsD1功能缺失型突变体,其中2株属于纯合突变类型。表型分析发现,OsD1突变体的内外稃出现显著的畸形发育,花粉粒的淀粉充实度降低且颖花不结实,初步推断水稻OsD1基因可能参与花器官的建成和发育。
OsD1, a gene associated with the development of rice panicle, was edited by the CRISPR/Cas9 genomeediting technology. Four mutant plants with defective genic function of OsD1 were obtained after the construction of site-directed editing vector, the transformation of rice callus, the identification of transgenic plants and the analysis of mutation types in the mutants. Among them, there were two homozygous mutants. The phenotypic analysis revealed that there existed significant deformities in palea and lemma of the mutants, the filling of starch in pollen grains of the mutants was poor and there were no seeds set on panicle in the mutants. Therefore, it is preliminarily concluded that the OsD1 gene might be relevant to the formation and development of floral organs in rice.
