响应面法优化酶法提取东北大马哈鱼中EPA的工艺研究 被引量:1

Study on the Extraction Process of EPA in Northeastern Salmon Using Enzyme Method Optimized by Response Surface

摘要 为了研究提取东北大马哈鱼中二十碳五烯酸(EPA)的最佳工艺,采用复合酶法,筛选复合酶的配比,并选取酶解温度、加酶量、p H、提取时间为影响因素,采用Box-Behnken响应面试验设计法,研究这4种因素对EPA产率和纯度的影响。结果表明:复合酶的最优配比为弱酸性脂肪酶、水解蛋白酶和多聚半乳糖醛酸酶质量比4︰3︰1;响应面法优化得到最优提取工艺:酶解温度42℃、加酶量1︰3、p H 6.5、提取时间5 h,每1.0 g样品加入复合酶0.02g。在此条件下EPA得率为23.2%,纯度经后续处理可达87.8%。 The optimum process for extraction of eicosapntemacnioc acid (EPA) in Northeastern salmon using compound enzyme method was studied. Firstly, screen the ratio of the compound enzyme, and then select enzyme hydrolysis temperature, enzyme amount, pH and extraction time as influential factors. The four factors effecting the yield and purity of EPA were studied by the Box-Behnken response surface test designing method. The results showed that the optimal ratio of the compound enzyme was the mass ratio of weak acid lipase, protease and polygalaeturonase 4 : 3 : 1. The optimum extraction conditions gotten by response surface method were as follows: the temperature was 42 ℃, the amount of ertzyme was 1 : 3, pH was 6.5, the extraction time was 5 h, and the amount of addition was the compound enzyme 0.02 g in 1.0 g sample. Under this condition, the yield of EPA was 23.2%, and the puritv was 87.8% after the subseauent treatment.
