摘要
目的探讨通络化瘀方对高级别脑胶质瘤化疗敏感性的影响。方法使用人脑胶质瘤细胞系U251为细胞模型。应用MTT试验及克隆形成试验检测不同组肿瘤细胞的活力和增殖能力。结果通络化瘀方治疗组肿瘤细胞活力及增殖能力较阴性对照组无明显变化,显示单用通络化瘀方对高级别脑胶质瘤无明显治疗作用。联合用药组及替莫唑胺组肿瘤细胞活力及增殖能力均低于阴性对照组;且联合用药组肿瘤细胞活力及增殖能力与替莫唑胺组比较明显降低。结论通络化瘀方在高级别脑胶质瘤中单用无明显治疗作用,而联合替莫唑胺治疗时可通过抑制肿瘤细胞的活力及增殖能力,从而促进其化疗敏感性。本研究为通络化瘀方应用于临床治疗脑胶质瘤提供理论基础。
Objective To investigate the effect of TongluoHuayu Decoction on chemotherapy sensitivity of high-grade glioma. Methods The U251 human glioma cell line was used as a cell model. MTT assay and colony formation assay were used to detect the viability and proliferation of tumor cells in different groups. Results The tumor cell viability and proliferation ability of TongluoHuayuDecoction group had no significant change compared with the negative control group, indicating that the use of TongluoHuayu Decoction alone has no obvious therapeutic effect on high-grade glioma. The tumor cell viability and proliferation ability of the combination group and temozolomide group were lower than that of the negative control group; and the tumor cell viability and proliferation ability of the combination group were significantly lower than that of the temozolomide monotherapy group. Conclusion TongluoHuayu Decoction has no obvious therapeutic effect in high-grade gliomas when used alone, and it can promote the chemosensitivity of tumor cells by inhibiting the viability and proliferation of tumor cells in combination with temozolomide. This study provides a theoretical basis for the clinical application of TongluoHuayu Decoction in the treatment of glioma.
