摘要
[目的]观察化痰通络汤对高脂血症合并脑缺血大鼠模型神经保护作用,探讨化痰通络汤通过抑制炎症反应途径抗脑缺血损伤的作用机制。[方法]建立高脂血症动物模型,以线栓法构建大鼠大脑中动脉栓塞复合缺血模型;采用酶联免疫法(ELISA)检测脑组织中COX-2、TNF-α、IL-1β的含量,免疫组化法测定大鼠脑组织中ICAM-1和VCAM-1的表达。[结果]中药治疗组与中药对照组相比较,神经功能评分降低,脑梗死体积缩小;脑组织匀浆中TNF-α、IL-1β、COX-2和脑组织中ICAM-1、VCAM-1的表达均减少。[结论]化痰通络汤可改善高脂血症合并脑缺血模型大鼠的神经功能缺失症状,缩小脑梗死体积,可能通过调节脑组织炎症反应、抑制炎症因子表达途径对抗缺血性脑损伤。
[Objective]To observe the neuroprotective effect of Huatan Tongluo Decoction in the Hyperlipidemia Merger of Cerebral Ischemia Model Rats, to explore the mechanism of the effect of Huatan Tongluo Decoction in preventing cerebral ischemic injury by inhibiting inflammatory response. [Methods]Building classic hyperlipidemia rat model with high fat diet,Constructing middle cerebral artery occlusion model; Detect the concentration of Cyclooxygenase-2( COX-2), Tumor Necrosis Factor( TNF-α) and Interleukin-1β( IL-1β) in serum between groups by ELISA. The expressions of intercellular adhesion molecule-1( ICAM-1), and vascular cell adhesion molecule-1( VCAM-1) in rat brain tissue were detected by immunohistochemistry.[Results]The treatment group was compared to the control group, Neurological scores were reduced. The volume of cerebral infarction shrank; The expression of TNF-α, IL-1β and COX-2 in the homogenation of the brain and the expression of ICAM-1 and VCAM-1 in brain tissue was decreased. [Conclusion ]The Huatan Tongluo Decoction can improve Hyperlipidemia Merger of Cerebral Ischemia Model Rats symptoms of neurological deficits, reduce infarct volume, it is possible to combat ischemic brain injury by regulating the inflammatory response of brain tissue and inhibiting the expression of inflammatory factors.
