摘要
目的了解杭州市水痘-带状疱疹病毒(Varicella-zoster virus, VZV)流行株基因型别特征。方法采集2012-2017年临床诊断水痘患者疱疹液,提取样本DNA,PCR扩增ORF 22、38、54、62基因片段。用单核苷酸多态性(Single nucleotide polymorphism, SNP)方法分析ORF 22基因片段,确定病毒基因型别;用限制性片段长度多态性(Restriction fragment length polymorphism, RFLP)技术分析ORF 38、54、62基因片段,区分野毒株和疫苗株。结果 23例临床诊断水痘患者的疱疹液中21例样本VZV核酸阳性。SNP分析结果显示,21例阳性VZV的基因型均属于clade 2。RFLP分析结果显示,21例阳性VZV均为野毒株(PstI~+BglI~+SmaI^-)。结论杭州市VZV流行株基因型别主要是clade 2,未发现疫苗株感染。
Objective To identify varicella-zoster virus(VZV) genotypes circulating in Hangzhou city. Methods We collected vesicle fluid samples from clinically-confirmed varicella cases during 2012-2017 for extracting DNA and amplifying fragments of ORF 22, 38, 54, and 62 by PCR. We used single nucleotide polymorphisms(SNPs) to analyze ORF 22 to identify genotypes and used restriction fragment length polymorphisms(RFLPs) to analyze ORF 38, 54, and 62 to differentiate wild strains from vaccine strains. Results Among 23 clinical-confirmed varicella cases, 21 were positive to VZV by PCR. SNP analysis showed that the genotypes of all positive VZV strains belonged to clade 2. RFLP analysis showed that all positive VZV strains were wild strains(PstI~+ BglI~+ SmaI^-). Conclusions The predominant genotype of VZV circulating in Hangzhou belonged to clade 2; infections by vaccine strains were not found.
