摘要
MYB转录因子是一类参与调控次生代谢和细胞形态、增强抗病性和应对生物和非生物胁迫反应的蛋白家族。本研究依据小兰屿蝴蝶兰(Phalaenopsis equestris)基因组和蝴蝶兰‘大辣椒’(Phalaenopsis‘Big Chili’)响应病毒胁迫转录组的测序结果,采用Pfam、HMMER、OrchidBase 3.0网站和Bioedit、MEGA 5.1、Jalview软件,对蝴蝶兰MYB家族基因进行了筛选和分类,分析了其基因类型、结构特点和系统进化关系,从小兰屿蝴蝶兰中筛选到152个MYB家族基因,其中R2/R3型6个、R2R3型138个、R1R2R3型3个。从大辣椒响应病毒胁迫转录组数据中共筛选得到55个MYB家族基因,其中有7个在响应不同时期差异表达明显,2个R2R3-MYB亚家族基因在感染病毒初期和携带病毒时期大幅上调,3个与小兰屿蝴蝶兰同源性较低。通过对小兰屿蝴蝶兰基因组和蝴蝶兰‘大辣椒’响应病毒胁迫时期MYB基因的筛选,为研究蝴蝶兰响应病毒关键MYB家族基因的分子功能提供了重要依据。
MYB TF is a protein family, which is involved in regulating secondary metabolism and cell morphology, enhancing disease resistance, and responsing to biotic and abiotic stresses. Based on the genome of Phalaenopsis equestris and transcriptome of the Phalaenopsis 'Big Chili' responsed to virus stress, this study screened and classified the MYB family gene in Phalaenopsis by the Pfam, HMMER, OrchidBase 3.0 and Bioedit, MEGA 5.1 and Jalview sotftware. Then we analyzed its gene types, structural features and phylogenetic relationships, as well as screened 152 MYB genes from Phalaenopsis equestris, including 6 R2/R3, 138 R2R3 and 3 R1R2R3.55 MYB genes were screened from transcriptome data of the Phalaenopsis 'Big Chili' response to virus stress, of which 7 were significantly differentially expressed at different response times. 2 R2R3-MYB genes were significantly up-regulated during the initial and symptoms stages after infection and 3 were lower homologous with Phalaenopsis equestris. The screen of the MYB gene in genome ofPhalaenopsis equestris and period of the Phalaenopsis Big Chili' response to virus stress might provide an important basis for studying the molecular function of MYB familygenes which was the key of Phalaenopsis response to virus.
